Protein phosphatase 2A(PP2A) is a serine / threonine protein phosphatase, It controls the phosphorylation of intracellular proteins, thereby regulating cell life activities. Such as: protein synthesis, cell signaling, cell cycle, apoptosis, metabolism and stress response, triggered by protein kinase signaling pathways regulating. Rapid progress is being made in understanding of PP2 A complex and its functions. Recent studies about PP2 A mainly associated with human disease changes, especially cancer. PP2 A is constituted of 3 subunits: a catalytic subunit, a scaffolding subunit, and a regulatory subunit. Alternating subunits can affect the incidence of malignant tumors. PP2 A inhibitors or direct activation of PP2 A drugs treatment for cancer has brought the dawn. At present, research on the regulation of PP2A-Aα gene has been some progress.In this thesis, I have explored the regulation of the scaffold subunit, Aα by the Transcription factors AP-2α and ETS-1. My results revealed the following.(1) AP-2α and ETS-1 are expressed in DU145 and LNCaP cells;(2) In DU145 and LNCaP cells, while ETS-1 seem to have little affect on PP2A-Aα, AP-2α negatively regulate the expression of PP2A-Aα;(3) AP-2α can bind to PP2A-Aα gene promoter;(4) In LNCaP, over expression of AP-2α down-regulates PP2A-Aα; In DU145 cells, silence of AP-2α up-regulates PP2A-Aα;Taken together, in LNCaP and DU145 cells, AP-2α negatively regulates expression of the gene coding for PP2A-Aα;...
