[Background]:Prostate cancer is one of the most common entity malignant tumor in United States and Europe, it is one of the leading causes of death among cancer patients.In recent years, the morbidity and mortality of prostate cancer in China all showed a trend of sustained growth, prostate cancer is becoming a serious threat on our men’s health.Patients with advanced prostate cancer are prone to bone metastases, resulting in poor quality of life and prognosis, but the transfer mechanism is unknown.Thus proved the metastasis of prostate cancer and look for effective drug is the problem of clinical treatment of prostate cancer.Nitidine chloride is extracted from the root of Zanthoxylum nitidum,it is a kind of pure natural alkaloid, our previous studies have shown that Nitidine chloride can inhibit inflammation effectively, also it can inhibit HIV/AIDS, function as antioxidant and inhibit the action of the fungus.And a series of recent studies have found that Nitidine chloride can inhibit proliferation and metastasis of breast cancer, kidney cancer, osteosarcoma and melanoma cells.This experiment is a preliminary assessment of Nitidine chloride effect on human prostate cancer cells PC-3 proliferation, invasion, and apoptosis,to assess it’s therapeutic value and discuss the possible regulatory mechanism. In this study, our data shows that Nitidine chloride inhibits prostate cancer cell invasion and migration. We also found Nitidine chloride play the role of inhibition of prostate cancer cell invasion and migration via Akt pathway.In short,these new findings has added new evidence for studying the role of Nitidine chloride inhibit cancer progression.Akt can suppress the progress of prostate cancer cells as a potential pathway.[Objective]:To evaluate the effect of Nitidine chloride on prostate cancer cells apoptosis and metastasis and it’s mechanism.[Materials and Methods]:1.We test the effect of Nitidine chloride on the proliferation of human prostate cancer cell PC-3 by MTT.2.Wound healing assay and Transwell were used to assess the effects of Nitidine chloride on the migration and invasiveness of prostate cancer cells.3.Flow cytometry assays were used to examine the changes in the apoptosis of PC-3 cells after Nitidine chloride treatment.4.The possible signaling molecules involved in this process were analyzed by immunoblot analysis.5.LY294002 was used to inhibit the activity of Akt pathway to further testing whether Nitidine chloride play a role in prostate cancer via Akt pathway.[Results]:1.Nitidine chloride can significantly inhibit the PC-3 cell proliferation and its inhibition has a time and dose dependent.2.Nitidine chloride can inhibit the PC-3 cell migration and invasion, and its inhibition has a dose dependent.3.Fluid cytology test shows PC-3 can be induced to apoptosis after Nitidine chloride treatment, and it’s effect on apoptosis has a dose dependent.4.Significant Increased the proportion of Bax/Bcl-2 (** P< 0.01) shows Nitidine chloride can induce the expression of apoptosis proteins5.LY294002 can significantly enhance Nitidine chloride not only induction of apoptosis of prostate cancer,but also inhibition of prostate cancer cell invasion.[Conclusion]:Nitidine chloride can suppress the proliferation, metastasis and induce the apoptosis of prostate cancer cells in vitro, it’s molecular mechanism to resist prostate cancer may be through the Akt pathway,suggesting its potential as a potential therapeutic agent for prostate cancer.
