ObjectiveAconite, which was first recorded in Shen Nong’s Herbal, has the effect of reviving the Yang for resuscitation, tonifying fire and helping yang, and eliminating cold to stop pain. However, aconite is itself toxic, which needs to be concocted before it can be used as medicine. Research shows that traditional processing methods are tedious, time consuming and energy consuming. Even worse, the loss of active ingredients is serious in preparing process, with uneven quality. Thus, it is difficult to guarantee the stability of the quality of Chinese medicine.Combined with previous production of medicinal pieces, task group has conducted further research on aconite about technology of high pressure steaming, high temperature baking, and microwaving. The best processing technology is determined by chemical composition. In order to prove scientifically whether new processing technology can completely replace the traditional, the project will give comprehensive evaluation, from the aspect of toxicology and pharmacodynamics, of modern processing technology of safety and effectiveness.Methods1. The study of literatureThis paper has analyzed and concluded on Aconite of processing technology, toxicological and pharmacological effects, which provides references for the formulation of feasible experiment scheme.2. Determination of the content of aconite and aconite decoction from different processing technologyFirstly, the same batch of aconite is processing by previously selected methods of high temperature, high pressure, microwave and that of black sheet. Secondly, HPLC method is respectively used to measure the alkaloids contents of aconite, its processed products and decoctions of processed products.3. Study on acute toxicity of aconite decoction of different processing technologyIt is carried out to evaluate the safety of four kinds of processed aconite through acute toxicity test.4. The establishment of mouse Yang deficiency model and experimental study on reviving the Yang for resuscitationMake Yang mold with hydrocortisone. Four kinds of processed aconite of different concentration decoction are given to Yang deficiency mice in order to evaluate its effect on reviving the Yang for resuscitation.5. Study on aconite decoction of different processing technology’s effect on Yang deficiency mice auricle microcirculation functionMake Yang mold with hydrocortisone. Four kinds of processed aconite of different concentration decoction are given to Yang deficiency mice in order to evaluate its effect on Yang deficiency mice auricle microcirculation function.6. Comparison of aconite decoction of different processing technology on mice cardiac functionWith isolated heart perfusion with Langendorff method, four kinds of processed aconite are given with the same volume of different concentrations of decoction, and to measure HR, LVSP, LVDP, and ±dp/dtmax. Results1. The results of content determination experiments show that the six major alkaloids and total alkaloids follow by high pressure sheets (referred to: GY), black smooth sheets (referred to:H), high temperature sheets (referred to:GW), and microwaving sheets (referred to:WB) from more to less.2. The acute toxicity results show that the maximum dosage of the four kinds of processed aconite is 48g crude drug · kg-1, which is equivalent to 192 times the clinical adult daily dosage(15g · 60kg-1), with no animal death.3. Reviving the Yang for resuscitation experiments show that comparing with plasma of the model group mice, the processed aconite of monkshood group can all reduce the plasma parameters of the Yang deficiency mice such as ET, c-GMP; and raise c-AMP/c-GMP、c-AMP. It can also increase the quality index of spleen and thymus. And having significant difference. Dosage between 0.75 to 7.04g/kg concentration range has good linear relationship with ET, C-GMP, c-AMP, spleen index and thymus index. And the drug treated mice have no significant difference between the different products.4. Compared with the model group mice, the diameter of capillary of drug treated mice is enlarged and the capillary network cross number is increased. Dosage between 0.75 to 5.63g/kg concentration range, with the increasing dosage, the difference between the two of diameter and network cross of capillary are gradually increasing.5. Dosage between 0.75 to 7.04g/kg concentration range, the four kinds of processed aconite can increase significantly in the isolated rat heart of HR, LVSP, LVEDP, and ± dp/dtmax with good linear relationship, and with obviously strengthening heart function. Each index of hearts of four kinds of processed aconite of monkshood group have no statistical difference. ConclusionStudys show that processed aconite decoction of 3 new processing technology has no obvious difference with traditional processing technology of the black sheet decoction in acute toxicity test, and function of reviving the Yang for resuscitation, Yang-warming and Vessels-activating, cardiac function. It states that processed aconite decoction of 3 new processing technology is safe and effective. And high pressure sheets has the same function with the traditional products in reviving the Yang for resuscitation. What’s more, the toxicity of the high pressure sheets is less than the traditional products. At the same time,it’s function of Yang-warming and Vessels-activating, cardiac function are also better than the traditional products. Therefore, it has important reference value for the improvement of the aconite processing technology, and provides the reference for the clinical use of aconite dosage, which provides an experimental basis for the further study of toxicology and pharmacodynamic aspects of the Aconite.
