Forsythia suspensa is the dried fruit of Forsythia suspensa(Thunb.) Vahl (Oleaceae). It has the function of Qingrejiedu and Xiaozhongsanjie. Modern pharmacological research indicate that Traditional Chinese medicine Forsythia is provided with antioxidation, antibiosis, anti-inflammatory, antivirus, anti liver injury and so on. Forsythiaside A is the basis of Forsythia, is provided with antibiosis, antivirus, relieve fever, analgesia, antioxidation and immunoregulation. This article researched the intervention of Forsythiaside A on carrageenan-induced paw edema and the metabolomic research of serum in rats from three levels whole, tissue and molecular with modern pharmacological research method, modern analytic technology and metabolomics. Expected to provide a scientific basis for the further development and utilization of the Forsythia resources and Forsythiaside A.1. Optimizing the separation and purification processes of forsythiaside A.Our group used the column chromatography twice to separate and purificate forsythiaside A rapidly with the preparative medium pressure liquid chromatography(Prep-MPLC). Firstly, we used polyamide as column material, intermediate products contained forsythiaside A more than 50%. Secondly, using MDS-5 column material to purify intermediate products, final products cantained forsythiaside A about 93%.2. Forsythiaside A act on the carrageenan induced paw edema in rats.By using the carrageenan induced inflammation rat paw edema, evaluation indicators are the temperature trends, swelling volume trends, the level of SOD, MDA, TNF-α, IL-2 and tissue sections. To research the anti-inflammatory, relieve fever and antioxidation of forsythiaside A. The results prove forsythiaside A has significant anti-inflammatory effect, antipyretic effect and antopxidation.3. Study on the metabolomics of serum in rats.This paper researched the effects of forsythiaside A, after carrageenan induced rat paw edema with metabolomics methods. The main research technique is nuclear magnetic resonance (’H-NMR) technology. To establish serum metabolic fingerprinting; analyse differences between the model groups and the others group by metabolite spectral and find out the biomarkers in serum; research the intervention on carrageenan induced rat paw edema from forsythiaside A; to discuss the metabolic pathways of forsythiaside A by comparing content change of the selected biomarkers. The study suggests carrageenan induced rat paw edema causes organism lipid metabolism, glycometabolism, amino acid metabolism are subject to different degrees of impact, and they can be effectively mitigated by forsythiaside A.
