Experimental Study Of Qi And Yin Deficiency Of Rat Pancreatic β Change In Type 2 Diabetes Mellitus And The Role Of Acupuncture Intervention

PurposesTo investigate the effect of acupuncture on apoptosis of pancreatic beta cells in pancreatic tissue in type 2 diabetic rats with Qi and Yin deficiency, PEPK, CHOP, Bcl-2, Bax protein and m RNA in changes under the action of acupuncture treatment, to provide a theoretical basis for clinical multi-way, multi-target therapy.Methods1.Select 60 male SD rats, and use high fat diet combined with intraperitoneal injection of STZ and stomach yin consumption concentrated decoction of Chinese Medicine, then make a copy of animal model of combination of Qi and Yin of deficiency type 2 diabetes syndrome.It is divided into 4 groups through the random number table method.The normal group has 11 rats,11 rats in the normal group,model group, acupuncture group and metformin group,with 12 rats in each group.Take Yishu, Zusanli,Qihai from the acupuncture group and treating continuously 6 weeks, metformin group were treated with metformin hydrochloride intragastric administration of 100mg/kg·d, it continues for six weeks, the model group and normal group were given equal volume of distilled water by intraperitoneal injection.2.During the process of experiment,record and observe the changes of rat diet, drinking water,urine volume every day,monitor rats’ weight and blood glucose regularly.Through the determination of expression of pancreatic tissues in PEPK, CHOP,Bcl-2,Bax protein and m RNA by Western Blot and RT-PCR, using HE staining method to observe the morphology of islet cells of rats in each experimental group.The experimental data were analyzed by SPSS19.0,using variance analysis,t test comparison of mean difference, P < 0.01 or P < 0.05 was statistically significant.Results1.After 6 weeks of treatment, rats in the acupuncture group of polyphagia,polydipsia, polyuria,emaciated,mental state,activity,sensibility,the glossiness of skin were improved than before.2.After 6 weeks of treatment,FBG of the acupuncture group and metformin group was significantly lower than that of model group(P<0.01),the difference was very significant. Acupuncture and metformin can reduce FBG of type 2 diabetic rats.3.After 6 weeks of treatment,PEPK,CHOP,Bax protein in pancreatic tissue in acupuncture group rat,compared with the model group,decreased(P<0.05). Compared with the model group,the acupuncture group rats’ s pancreatic tissue with Bcl-2 protein increased(P<0.05).The effect of Metformin treatment group is inferior to acupuncture group.4.After 6 weeks of treatment,PEPKm RNA,CHOPm RNA,Baxm RNA expression in pancreatic tissue of rats in the acupuncture group,compared with model group, decreased significantly(P<0.01).Compared with the model group,pancreatic tissue Bcl-2 gene expression in rats of acupuncture group increased(P<0.05).The effect of acupuncture group and metformin group treatment is about the same.5.After 6 weeks of treatment,from the morphological observation,the rats in the model group compared with normal group,the islet structure was damaged obviously,the number of valid islet cells was significantly reduced.The acupuncture group, metformin group,the number islet cells and islet cells structure was significantly better than the model group.Conclusions1.Use the improved method to establish the Qi and Yin deficiency rat model of type 2 diabetes,basically reflects the human Qi and Qin deficiency symptoms characteristic of type 2 diabetes mellitus,which have the characteristics of Qi and Qin deficiency syndrome.2.Type 2 diabetic rats after acupuncture treatment significantly improved the symptoms of Qi and yin deficiency,and was superior to metformin group; acupuncture and metformin can reduce the fasting blood glucose in type 2 diabetic rats;The determination of acupuncture can regulate the expression of PEPKm RNA, CHOPm RNA,Bcl-2m RNA,Baxm RNA,PEPK,CHOP protein expression wassignificantly decreased after treatment,and better than metformin group,acupuncture can regulate up the Bcl-2 protein,and the regulate down the Bax protein.Considering the deficiency of Qi and Yin symptoms in type 2 diabetic rats may improve with the m RNA and protein regulation,so as to reduce the apoptosis of islet beta cells,has the reference significance to the clinical research.
