Objective: Observation of Yang Gan Yi Shui granule 12 week old SHR rats(Alb) urinary albumin / creatinine(Cr) ratio(ACR) and blood Scr, BUN content and renal pathological changes; and the impact of renal cortex in SHR rats TGF beta 1, PCNA,p27 and p57 m RNA and protein expression and beta 1, PCNA, TGF p27 and p57 m RNA and protein expression. In order to elucidate Yang Gan Yi Shui Granule on early podocyte function in hypertension intervention and the mechanism, to provide experimental evidence for treatment of early renal damage in essential hypertension in glomerular sclerosis, for the improvement of Yang Gan Yi Shui granule.Methods: A total of 64 SHR rats were randomly divided into 4 groups, respectively,Yanggan Yishui Granule high dose group, Yanggan Yishui Granule low dose group,western medicine group, model group, groups of average, 16, and 16 WKY rats as control group. Adaptive feeding one week before the experiment, observation of the physiological status of rats, using picric acid marker number. Determination of blood pressure meter detecting each stage of rat type 1.RBP1 value of heart rate in rat tail artery blood pressure; 2.The experiment of automatic biochemical analyzer was used to detect the changes of serum creatinine, urea nitrogen index of rats; 3.observation of sixth weeks,were treated with drugs for 12 weeks after the intervention of pathological changes in kidney of rats in each group with each group; 4.rat kidney tissue of experimental detection technology in RT-QPCR after TGF beta 1, PCNA, p27 and p57 m RNA expression by Western; 5.Western blot rats kidney TGF beta 1, PCNA, p27 and p57 protein expression after the test; 6.SPSS 18 software package was used for statistical processing.Results:(1) The behavior of rats after drug intervention activities, a series of changes:Yang Gan Yi Shui granule high dose group to improve the situation close to the normal group; with Yang Gan Yi Shui granule high dose group to intervention, eyeball inspontaneously hypertensive rats by dark red to pale red, hot tempered by gradually become docile, easy to catch, hair color is moist, fight bite phenomenon no longer appear between the treatment groups before the experiment; the blood pressure in SHR rats compared with the normal group of WKY rats(P < 0.01); during the experiment of SHR rats in model groups rats with age, blood pressure increased gradually, and the western medicine group, high Yang Gan Yi Shui granule dose group, low dose group of Yang Gan Yi Shui granule rat blood pressure was decreased slowly, and three in the treatment group blood pressure levels are very close, but the high dose of Chinese medicine group decreased greatly compared with other groups.(2) before and after treatment SHR in model group rats blood Scr, BUN was significantly higher than the normal group WKY rats(P < 0.01); after the treatment, each treatment group decreased significantly SHR in rat blood Scr, BUN levels(P < 0.01), but no significant difference between the three groups(P >0.05).(3) before and after treatment in each group of SHR rats urine ACR was higher than that in the normal group WKY rats(P < 0.01); after treatment, the treatment group SHR rat urine ACR were decreased, and the model group compared with significant difference(P < 0.01); low dose of Chinese medicine group and Western Medicine group compared statistically significant(P > 0.05), and high dose of Chinese medicine group and low dose traditional Chinese medicine group and benazepril group compared with statistical significance(P < 0.05).(4)after treatment in each group SHR rats compared to the normal group WKYin rats with glomerulosclerosis is more obvious, and the treatment groups, compared with before treatment reduced the degree of glomerular sclerosis.(5) model group of renal cortical TGF beta 1m RNA relative expression was significantly higher than normal group(P<0.01); after treatment, Yanggan Yishui Granule high, low dose group, western medicine group than those in the model group of renal cortical TGF beta 1m RNA relative expression quantity decreased(P<0.05; P<0.01); Yanggan Yishui Granule high dose group compared with Yanggan Yishui particles of low dose group of renal cortical TGF beta 1m RNA relative expression significantly decreased(P<0.05); model group,renal cortical p27 P57 m RNA relative expression level was significantly higher than normal group(P<0.01); after treatment, Yanggan Yishui Granule high dose group,western medicine group than in the low dose group of Yang Gan Yi Shui granule renal cortex p27, P57 m RNA relative low expression level(P<0.05); model group of renal cortical PCNAm RNA relative expression was significantly lower than that in normal group(P<0.01), after treatment, Yanggan Yishui Granule high dose group, western medicine group with Yanggan Yishui Granule low dose group of renal cortical PCNAm RNA relative expression quantity increased(P<0.01).(6) model group of renal cortical TGF beta 1 protein expression levels were significantly higher than that of the normal group(P<0.01); after the treatment, the expression level of Yang Gan Yi Shui granule high dose group, western medicine group than those in the model group of renal cortical TGF beta 1 protein decreased significantly(P<0.05; P<0.01); Yanggan Yishui Granule high dose group than in the liver and water feed particles of low dose group of renal cortical TGF expression levels of 1 protein beta decreased significantly(P<0.01);the expression of renal cortex p27 model group, P57 protein levels were significantly higher than those in the normal group(P<0.01); after treatment, Yanggan Yishui Granule high dose group than in low dose group of Yang Gan Yi Shui granule in renal cortex p27, the expression level of P57 protein reduce(P<0.05; P<0.01); the expression level of PCNA protein in renal cortex of model group was significantly lower than that in normal group(P<0.01), after treatment, Yanggan Yishui Granule high dose group,western medicine group with Yanggan Yishui Granule low dose group of renal cortical PCNA protein expression of water Ping Zenggao(P<0.05).(2) Conclusion: 1. Yang Gan Yi Shui granule can significantly reduce SHR rat urinary ACR ratio, serum creatinine, blood urea nitrogen, blood pressure, and improve the renal damage. 2. Yang Gan Yi Shui granule by regulating TGF beta 1 expression to regulate PCNA, p27, p57 protein expression, thereby regulating the proliferation and apoptosis of podocytes and inhibition of renal fibrosis and slow the progression ofglomerulosclerosis and maintain normal podocyte morphology and structure, so as to achieve the purpose of protecting the kidney.
