The Study On The Relationship Between TCM Syndromes Of Chronic Heart Failure And The Tongue Flora Compositions

ObjectiveTo investigate the composition differences of tongue coating flora between chronic heart failure patients and healthy people, and between different TCM syndrome types of chronic heart failure patients.MethodsIncluded the patients of qi and yang deficiency blood stasis fluid retention (yang deficiency group)、qi and yin deficiency blood stasis fluid retention (yin deficiency group) and healthy people from physical examination center. All of the cases are from in-patient department of the first affiliated hospital of Guangzhou university of TCM and Guangdong hospital of TCM. To note the people’sgeneral information, personal history, clinical date, Xiang of tongue and so on. Collect their tongue flora samples, DNA extraction, and sequence by Illumina, then analyze the biological information. Get the flora bacteria’s abundance and diversity, and then analyze the differences among groups.Result1.This study Screened 60 patients diagnosed with chronic heart failure, 18 cases of them has been using antibiotics in hospital, the actual choosed into the yang deficiency group was 19 cases, yin deficiency group was 23 cases; At the same time This study Screened 30 cases of healthy people, among them 1 case had upper respiratory tract infection in 1 week, actually included in the healthy group was 29 cases.2. Three groups of patients have no significant differences in gender. Yang deficiency group and yin deficiency group of chronic heart failure patients have no significant difference in age, smoking, drinking, heart function grading.3. Yang deficiency group of tongue color is given priority to with light and dark, Yin deficiency group of tongue color is given priority to with dark red and red, healthy people group of tongue color is given priority to with pink. Yang deficiency group’s tongue coating quality is given priority to with greasy or mild greasy, Yin deficiency group’s tongue coating quality is given priority to with less coating or thin coating, healthy people group’s tongue coating is given priority to with thin coating.4. The sequence quality has reached the standard. There are 4092353 pairs of reads left after filtering. No.80 sample was removed due to the low sequence reads.5. In 70 samples sequencing, measured 1 super kingdom,12 phyla,21 classes, 32 orders,54 families and 89 genera.6. The rarefaction curve and Shannon-Wiener curve response the sequencing depth is enough.7. The PCA analysis, community heatmap, and Weighted UniFrac analysis shows that the abundance and diversity of the flora bacteria among three groups is no significant difference. But there are different variation trends of the flora bacteria composition among three groups. The difference between chronic heart failure in the two groups of tongue coating flora is that Prevotella^ Veillonella content is reduced, at the same time Neisseria content increased. In CHF of qi and yang deficiency blood stasis fluid retention, the Streptococcus content was decreased and Fusobacterium content was rising, compareing to the qi and yin deficiency blood stasis fluid retention group on flora in tongue coating.ConclusionThere are no significant difference of the flora bacteria abundance and diversity among yang deficiency group, yin deficiency group and healthy group. But on the flora bacteria composition has different variation trends. The difference between chronic heart failure in the two groups of tongue coating flora is that Prevotella、Veillonella content is reduced, at the same time Neisseria content increased. In CHF of qi and yang deficiency blood stasis fluid retention, the Streptococcus content was decreased and Fusobacterium content was rising, compareing to the qi and yin deficiency blood stasis fluid retention group on flora in tongue coating.
