Objective:The aim of this study is to investigate the predictive values of matrix metalloproteinase-2 (MMP-2) and matrix metalloproteinase-9 (MMP-9) in plasma to the injury of the middle layer of aorta in CHD patients with hypertension, and to study the relationship between the position of injury in the middle layer of aorta and the expression of MMP-2 and MMP-9.Method:Thirty random CHD patients with hypertension in TED A international cardiovascular hospital were enrolled in our study. This study group consisted of 15 males and 15 females,aged from 48 to 76 years(mean:60.5± 8.2),all patients were treated by coronary artery bypass grafting surgery. Blood sample was collected before surgery, while aortic tissue was gathered through the surgery. Pathological staining was performed in parts of aortic tissue of these patients. And the concentration of MMP-2 and MMP-9 in plasma or in the homogenate of aortic tissue were measured with the method of ELISA. All analyses were performed by SPSS 17.0,with P<0.05 was considered significant.Result:(1) MMP-9 in plasma had a trend of correlation to the MMP-9 in the homogenate of aortic tissue (r=0.433,P=0.061). (2) MMP-2 in plasma did not correlate to the MMP-2 in the homogenate of aortic tissue (r=0.033,P>0.05). (3) Pathological staining showed that the injury degree in the middle layer of aorta near intima was more serious than the injury degree in the position near adventitia. (4) IHC staining showed that MMP-2 expressed abundantly in the middle layer of aorta.And,MMP-2 mainly localized in the pathological progress areas of middle layer.Conclusion:In the CHD patients with hypertension,we concluded that:(1) MMP-9 in plasma possibly can be used to predict the injury of the middle layer of aorta, which still need further study to confirm. (2) MMP-2 in plasma may not be used to predict the injury of the middle layer of aorta. (3) The injury degree in the middle layer of aorta near intima is more serious than the injury degree in the position near adventitia. (4) The important role that MMP-2 plays might be in terms of the degradation of extracellular matrix in the middle layer of aorta.
