Objective:To study the toxin effect of Pseudomonas aeruginosa on lifespan, reproduction,growth development,fat storege of Caenorhaditis elegans and test the expression of cyp35 family genes to discuss pathogenic mechanism of Pseudomonas aeruginosa on C. elegans P450 metabolic signaling pathway, we measured the Pseudomonas aeruginosa’s density and concentration of toxic emisssion to definite killing factors.Method:(1)Using the C. elegans-pseudomonas aeruginosa infection based model for studying host pathogen interactions that is genetically tractable from both the host and pathogen. Observe the changes including lifespan, body length, motivation and eggs-laying number of C. elegans after being infected by pseudomonas aeruginosa cultivated by different medium.(2)To test the expression of cyp35 genes cytochromeP450 including cyp35Al, cyp35A2, cyp35A5 and cyp35C1 genes by RT-PCR.(3)Oil Red O was used to quantitata fat content of C. elegans in the 24 wells-plates.(4)Measurements were made of the density and different toxicant concentration of pseudomonas aeruginosa cultivated by different liquid culture at different wavelength.Results:(l)The difference in survival time and eggs-laying number after being infected by pseudomonas aeruginosa statistically significant(p<0.05); C. elegans was completely deprived of motivation ability on the TSA medium and the movement flexibility has reduced, germinate and growth was inhibited by pseudomonas aeruginosa; C. elegans body length on the LB medium was the shortest, the flexibility of motivation has reduced.(2)The gene expression of cyp35A1, cyp35A2, cyp35A5 and cyp35C1,was up-regulated in different degree among three group C. elegans infected by pseudomonas aeruginosa.(3)C. elegans’s fat storege was reduced after being infected by pseudomonas aeruginosa.conclusion:Pseudomonas aeruginosa ATCC27853 could lead to slow and stunted gr-owth, a clear body with sluggish movement, eggs-laying number-reduction, and some embryonic lethality and reduced fat storege and lifespan on C. elegans, the toxin emission of psedomonas aeruginosa lead to the expresssion of cyp35 genes(cyp35A1,cyp35A2,cyp35A5, and cyp35C1) up-regulated. Pseudomonas aeruginosa could reduce fat storege in C.elegans. QS system and bacterial boifilm played a regulatary role in the virulence of pseudomonas aeruginosa to C. elegans.
