Objective To detect the mechanism of NM23-H2 inhibiting invasion and migration in gastric cancer cells.Methods1. We used the Western blot method to identify expression of NM23-H2 in BGC-823 cells which were transfected NM23-H2.2. Cell morphology was observed with inversion microscope.3. The levels of E-cadherin and vimentin were determined by Western blot.4. The expression of LC3II was explored by Western blot.5. The ultrastructure of autophagic vacuole was observed by TEM.6. Autophagosome was observed by the MDC staining immunofluorescence.7. The optimal concentration of chloroquine was confirmed by MTT, using the Western blot to explore the expression of E-cadherin, vimentin and LC3II inhibiting antophagy by CQ.8. We used Transwell method to confirm the influence of NM23-H2 on invasion and migration in BGC-823 cells.Results1. The expression of NM23-H2 was increased in BGC-NM23H2 cells (P<0.01).2. We used 10ng/ml TGF-β1 to induce EMT, the expression of E-cadherin was increased and vimentin was decresed in BGC-NM23H2 cells (P《0.001).3. The expression of LC3II was increased obviously in BGC-NM23H2 cells. Transmission electron microscope (TEM) and MDC staining results were identified with the result of Western blot.4. We used chloroquine to inhibit autophagy, the expression of E-cadherin downregulated and vimentin upregulated (P<0.01) in BGC-NM23H2 cells.5. The invasion and migration were inhibited in BGC-NM23H2 cells (P<0.01). After inhibiting autophagy, the invasion and migration were increased in BGC-NM23H2 cells (P<0.01).Conclusions1. NM23-H2 can inhibit EMT in BGC-823 cells.2. NM23-H2 induces autophagy in BGC-823 cells.3. NM23-H2 inhibits EMT through inducing autophagy in BGC-823 cells.4. NM23-H2 inhibits the invasion and migration through inducing autophagy to inhibit EMT in BGC-823 cells.
