Objective The vast majority of the Malignant Tumor of Oral and Maxillofacial Region is oral squamous cell carcinoma. Now, OSCC is now popular and frequently.This has resulted in serious harm to the patient’s life and the quality of Life. Late metastasis, recurrence is the most intractable problems for clinicians and main reasons to death. Researchers investigate the metastasis mechanism of oral squamous cell for many years. In recent years, we find that voltage-gated sodiu channel expressed in some cancers. Such as prostate cancer, breast cancer, non-small cell lung cancer, ovarian cancer, cervical cancer, etc. To investigate the expression of Nav1.6 in oral squamous cell carcinoma and the relationship between Nav1.6 and the development of oral squamous cell carcinoma.Methods Surgical tissue specimens from 67 patients with OSCC who underwent surgical treatment at the First Affiliated Hospital Of Anhui Medical University, the histological diagnosis was confirmed by an experienced pathologist. And non-cancer tissue from gum and mucocele. Then Immunohistochemistry, Real-time Quantitative PCR were used to detect the expression of Nav1.6 in carcinoma and normal human oral tissue. Then analysis the results by using instrument. But, there is no specific and sensitive tumor marker can accurately diagnose OSCC.Results The results of Real-time Quantitative PCR revealed that the expression ofNav1.6 in oral squamous cell carcinoma increased significantly in comparison with normal oral tissue,the higher the degree of tumor differentiation, the less the expression of Nav1.6. The average relative expression levels of 16 non-cancer tissue is 1.00±0.14. The average relative expression levels of well-differentiated squamous cell carcinoma is 2.31±0.52. The average relative expression levels of moderately differentiated squamous cell carcinoma is 4.46±0.58. The average relative expression levels of poorly differentiated squamous cell carcinoma is 8.86±1.63. Immunohistochemistry revealed that the expression of Nav1.6 located in the cell membrane and cytoplasm, especially in the cancer nest, keratin pearls.The expression levels in OSCC tissues was significantly higher than non-cancer tissue.This has nothing to do with age, sex, the site of tumor and the metastasis of lymph nodes. The Real-time Quantitative PCR result revealed that the mean relativity expression of Nav1.6 m RNA in tumor was increased 5.24 times(p<0.05) compared with the normal oral tissue.Conclusion This experiment confirmed that there are plenty of Nav1.6 expression in oral squamous cell carcinoma. The higher the degree of tumor differentiation,the less the expression of Nav1.6.The up-regulated level of Nav1.6 m RNA in oral squamous cell carcinoma tissue are significantly associated with the pathogenesis of OSCC. Nav1.6 is likely to become a new tumor markers for studying OSCC.
