Background:The research on changes of Human pathology and Physiology in the surrounding of microgravity is taken much more attentios than before. skeletal system is one of important part of human body, of which the osteoblast is helpful to its stabilization. Pathologic and Physiological functions of astronauts have many changes like amyotrophy, hypoimmunity, loss of sclerotin and so on in microgravity. And the changes on skeletal system are obvious, include the loss of sclerotin, deviantbone micro architecture and osteoporosis. Under the circumstances, more useful medicine to cure or prevent is in urgent need in clinic.Research method:To research the effect of osteoblast biology of the rat calvaria bone in microgravity.1. We set a scientific model for the growing of osteoblast in microgravity, and the cells are from the new born(in 24 hours) rats’ cranium. 2. In this model, the oscillating has frequences from 5 to 90 Hz, and the cycle is 10 minutes. The type of oscillator is BOSE 3100 biomechanics testing machine, and the microgravity is 0.5g. 3. Record the influences of proliferative effect and functional differentiation of osteoblast in microgravity by MTT chromatometry and ALP detection kits.4. Probe the influences of proliferation and differentiation of rats’ craniumosteoblast in variable frequency vibration in microgravity.Result:1. Separate and incubate the new born(in 24 hours) rats’ cranium osteoblasts by enzyme digestion;found a scientific model of culture in vitro Of the new born(in 24 hours) rats’ cranium osteoblasts in Two-way and Multi-Sample in microgravity. 2. We find proliferation and differentiation of rats’ cranium osteoblast in variable frequency vibration in microgravity are inhibited by MTT chromatometry and ALP detection kits in microgravity. 3. The variable frequency(90Hz and 5-90Hz) vibration can promote the proliferation of osteoblast by MTT chromatometry in microgravity. 4.The variable frequency(45Hz and 5-90Hz) vibration can promote the differentiation of osteoblast by ALP detection kits in microgravity.Conclusion:1. The functions of proliferation and differentiation of rats’ craniumosteoblast in microgravity are inhibited. 2. The variable frequency vibration can protect the functions of proliferation and differentiation of osteoblast in microgravity. 3. This research provide the experimental and theoretical basis for that mechanical vibration can cure and prevent the loss of bone mass in microgravity.
