Objective:Observation of didangqiguirecipe on diabetes mellitus(DM) insulin resistance(IR) in rat serum free fatty acid(FFA) and adiponectin(ADPN) content and the influence on the structure of the islet beta cells and to explore the mechanism of IR and glucose and lipid metabolism and internal relations of the party and further provide more powerful experimental basis for didangqiguirecipe prevention and treatment of diabetes.Method:Will be 80 SD rats were randomly selected 10 rabbits as blank control group, the remaining 70 only given high sugar, high fat diet, in the eighth week 7 days 20 when begin fasting. The next day as early as 8 to be a single intraperitoneal injection of streptozotocin(STZ)(STZ(placed in an ice bath) 35 mg / kg, 72 h after measured blood glucose and measuring the random blood glucose, urine glucose, 16.7mmol/l glucose is more than or equal to, urine(3 +) or(4 +) [1]. Will make the successful model of the rats were randomly divided into 5 groups, the model control group, Didang Qigui Decoction high dose group(Chinese medicine group, a large number of), didangqiguirecipe conventional dose group(hereinafter referred to as the traditional Chinese medicine constant group), hydrochloric acid than pioglitazone group(referred to as the Western medicine group, Didang Qigui Decoction by hydrochloric acid pioglitazone group(Western medicine group). Intervention for 8 weeks, at the end of the 8th week by enzyme linked immunosorbent method(ELISA) for detection of rat serum ADPN levels, with a colorimetric method to determine the content of serum FFA,(admin) SP immunohistochemical method to detect the rat serum insulin(fins) was used, with light microscope observation of changes of rat pancreatic beta cell structure.Result : Compared with the model group, each treatment group rats polydipsia, polyuria, and other symptoms were improved, weight: the intervention group(Chinese medicine group in addition to a large number of) the rest of body weight of rats in each group were increased than before, compared with the model group changed significantly(P<0.05); blood glucose: each treatment group(except in a large number of drug group) rats blood glucose were lower than before(P<0.05), FFA, FINS; serum: the intervention group rats FFA, FINS content compared with the model group have different degrees of reduction, especially in traditional Chinese medicine group, western medicine group constant changes, the most obvious(P<0.01): ADPN; the intervention group the content of ADPN increased in varying degrees compared with the model group, especially the traditional Chinese medicine group and Western medicine group. The most obvious(P<0.01).Conclusion:Didang Qigui Recipe in a certain extent improve diabetic rats of insulin resistance, reduce blood sugar, correct the disorder of lipid metabolism, especially with its constant effect is more obvious. The mechanism probably is by decreasing the content of serum FFA, increased the content of serum ADPN and reduce the content of serum fins, increase peripheral tissue glucose utilization, enhanced insulin sensitivity, delay and improve the islet beta cell apoptosis, so as to achieve the purpose of prevention and treatment of diabetes.
