PurposeTo observe the proliferation and the expression of hyaluronan synthase(HAS) mRNA of the orbital fibroblasts from health control and the patients of thyroid-associated ophthalmopathy(TAO),which added sCD40L.Explore the role of sCD40L in the pathogenesis of TAO.Methodsorbital fibroblasts obtained from health control of 2 person and 5 patients with TAO. The effects of sCD40L on the proliferation of the fibroblasts were compared with MTS colorimetry assays. The effects of sCD40L on the expression of HAS mRNA in the fibroblast was examined by RT-PCR.Results(1) The orbital fibroblasts were cultured successfully. (2) More than 25 ng/ml soluble CD40L induced the proliferation of the orbital fibroblasts,and was it was in a time-and dose-dependent manner(3)The expression of HAS 1 and HAS2 mRNA are lower than the up-regulated expression of HAS3 mRNA. The HAS1,HAS2 and HAS3 of Normal orbital fibroblasts had no obvious change in various concentration.ConclusionsOur observations suggest that sCD40L may play a role in the pathogenesis of TAO.And the sCD40L is not only have impact to the proliferation of orbital fibroblasts,but to expression of HAS mRNA.It promotes the TAO orbital fibroblasts synthesis and secretion of patients with HA, thereby promoting the development of TAO.
