| Objective The study was designed to construct Lenti-mi R-210-Luciferase, and detect angiogenic factors HIF-1α and VEGF expression in human periodontal ligament stem cells(h PDLSC) transduced by Lenti-mi R-210-Luciferase.Methods Human PDLSCs were isolated and cultured in vitro, and Lenti-mi R-集210-Luciferase(control group, Lenti-Lac Z-Luciferase) was constructed. After lentiviral titer was detected, human PDLSCs was transduced by Lenti-mi R-210-Luciferase. The analysis of HIF-1α and VEGF expression was done with q PCR and the immunohistochemistry examination.Results After Lenti-mi R-210-Luciferase was transduced to human PDLSCs at 0 d, 1 d, 4 d, 7 d and 14 d, the results of q PCR showed that the over-expression of HIF-1α and VEGF was detected on 4 d, HIF-1α expression was highest on 7 d, and remained at a higher level on 14 d; VEGF expression still had a rising trend until 14 d. Immunohistochemical results showed that after human PDLSCs was transduced by Lenti-mi R-210-Luciferase, PDLSCs were positive for HIF-1α and VEGF antibody, control group was negative.Conclusion We successfully constructed the Lenti-mi R-210-Luciferase, and prove that mi R-210 can promote human PDLSCs the differentiation of angiogenesis. |
