| Objective:Diabetic macroangiopathy is not only one of the vascular chronic complications of diabetes, but also one of the important causes of the death of DM.Therefore,proactively preventing and treating early diabetic macroangiopathy have a big significance. Yi Qi Yang Yin Huo Xue recipe is in common use in the treatment of diabetic macroangiopathy. This experiment built diabetic rats model by injecting Streptozotocin(STZ), then give the Yi Qi Yang Yin Huo Xue recipe to the diabetic rats, and surveyed the diabetic rats’ changes on weight, general behavior, glucose,blood lipid, blood vessel endothelium form and the NF-κB.In the last, discussing the possible function mechanism on prevention and treation diabetic macroangiopathy by using the prescription and the effect of this prescription.Methods:In this experiment we select fifty male clean Wistar rats, with the Weight about 230 g ~ 230 g.Firstly we feed these rats by ordinary diet for four days,then randomly select 6 rats as normal group(NG), other fiftyfour rats were injected streptozotocin(STZ) in abdomen according to 60mg/kg,which was dissolved into 1% buffer solution of citricacid of STZ. The NG were given corresponding 1% citric acid, sodium citrate solution injection as the same volume as the model bulit group. After seventy-two hours, the tail vein of rats collected, measured blood glucose more than 16.7mmol/L,in the same time water intake and urine output increased significantly in rats,these show the successful model. The successful model of the 44 rats were randomly divided into model group(MG), Chinese medicine group( Yi Qi Yang Yin Huo Xue’s group ZG), Western medicine group(simvastatin group,XG), Chinese medicine and western medicine group( yiqiyangyinhuoxue and simvastatin group,ZXG), each group of 11.After a week, The rats of ZG were given the concentrated liquor of yiqiyangyinhuoxuefang,according to 10 m L/kg,the XG’s rats were given the simvastatin,according to 0.33 mg / kg,the ZXG’s rats were given yiqiyangyinhuoxuefang equal to ZG’s at morning and simvastatin equal to XG’s at afternoon,the NG’s rats and the MG’s rats were given the corresponding quantity’s physiological saline by gavage.All the rats were given drugs once a day for 12 weeks. All rats were water-deprivation and fasting for 12 hours after the last lavage,then these rats were anesthetized by injecting 2%napental into abdomen,and broken to collect blood to test plasma total cholesterol(TC), blood glucose(BG),high density lipoprotein cholesterol(HDL) and low density lipoprotein cholesterol(LDL). In the last, get the specimen,and observe the change of pathomorphism of macrovascular endothelial under light microscope, immunohistochemical to measuring thoracic aorta endothelium vascular of NF-κB.Results: 1 The general situation of rats NG’s rats drinking, eating and urine were regular. The weight of rats had significant growth.They had a good mental state, color luster,activities freely. Rats in MG were more drink(typical symptom), polyuria(typical symptom), polyphagia(typical symptom), emaciation(typical symptom), color yellow, poor mental state, decreased activity. Part of the rats,in the progress of the experiment, appear a series of diabetes complications, such as infection, limb edema,etc. The performance of the treatment groups was worse than NG,but better than MG.In MG and XG,eventually death number of rats respectively2,2.The analysis of the main cause of death is skin infection or biting each other.In ZG, death number of rats is 1,becase of filling the stomach. 2 Blood sugar(BG) change. The MG and the treatment groups(ZG,XG,ZXG) rats’ BG were higher than the NG,the difference was statistically significant(P<0.05),but the XG was not of significant differences, comparing with the MG(P>0.05).Comparing with MG,the BG of ZG and ZXG decrease,the difference was statistically significant(P<0.05), but the ZG was not of significant differences, comparing with the ZXG(P>0.05). 3 Observe blood lipid(TC,LDL,HDL). The MG and the treatment groups(ZG,XG,ZXG) rats’ TC were higher than the NG,the difference was statistically significant(P<0.05),but the ZG was not of significant differences,comparing with the MG(P>0.05). Comparing with MG,the TC of XG and ZXG decrease, the difference was statistically significant(P <0.05), but the XG was not of significant differences comparing with the ZXG(P>0.05). The MG and the treatment groups(ZG,XG,ZXG) rats’ LDL were higher than the NG,the difference was statistically significant(P<0.05),but the ZG was not of significant differences,comparing with the MG(P>0.05). Comparing with M G,the LDL of XG and ZXG decrease,the difference was statistically significant(P<0.05), the ZXG was lower than the ZXG,the difference was statistically significant(p<0.05). The MG and the treatment groups(ZG,XG,ZXG) rats’ HDL were lower than the NG,the difference was statistically significant(P<0.05),and the treatment groups(ZG,XG,ZXG) rats’ HDL were higher than the MG,the difference was statistically significant(P<0.05). Comparison between three treatment groups, the HDL change significantly, the difference was statistically significant(P<0.05). 4 Observation of pathological morphology on macrovascular endothelial tissue. After HE staining, NG cells display texture clear, shape was normal, cell cytoplasm coloring uniformity, aortic endothelial cells lining flat, smooth, in the film smooth muscle cell and elastic plate and present a parallel arrangement. MG of rat aortic intima thickening of the limitations, endometrial local protuberance, membrane shallow of smooth muscle cells visible hyperplasia, arrangement disorder, elastic plate arrangement irregular wavy, rarely appear rupture, separation. The treatment groups’ sform of organization was better than the MG.,but worse than the NG. 5 Expression of NF-κB in rats’ vascular. Comparing with the NG, MG and the treatment group(ZG,XG,ZXG)rats thoracic aorta vascular NF-κB is obviously higher(P<0.05).Comparing with MG,the NF-κB of ZG and XG and ZXG decreased, the difference was statistically significant(P<0.05), Comparison between three treatment groups, the NF-κB reduce significantly, the difference was statistically significant(P<0.05).Conclusions: 1 The Yi Qi Yang Yin Huo Xue recipe could improve the general condition of the diabetic rats, because it could reduce blood lipid,reduce blood glucose, improve diabetes rats’ macrovascular endothelial tissue pathological form. 2 The possible function mechanism of prevention and treation about Yi Qiyangyinhuoxuefang on diabetic macroangiopathy should be owing to its improvement of the blood fat and blood glucose, restrain the expression levels of NF-κB. |
