Objective: In recent years,with the improvement of living standards and continuous intake of high-fat diet,the incidence of metabolic syndrome increased every year which is formed by the merger of various metabolic diseases(such as central obesity,diabetes or impaired glucose tolerance, hypertension,dyslipidemia and hyperuricemia).Moreover,in the condition of the prevalence of increasingly younger age,Another hazard of metabolic syndrome appears day by day that is erectile dysfunction. It refers to the penis cannot achieve and(or) maintain an erection sufficient for satisfactory sexual life. Numerous epidemiological and clinical researches show that there is a close connection between metabolic syndrome and erectile dysfunction. In recent years,the relationship between erectile dysfunction and metabolic syndrome has gradually become a hotspot.But the domestic research and reports are limited to overview.In this study, we would discuss the impact and mechanism of metabolic syndrome on erectile function of rats combining with the indicators to provide reference for the future of diagnosis and treatment of MS-ED.Methods: There are 20 healthy male Wistar rats which were randomly divided into HFD group(n=10) and NPD group(n=10).Model group was given the 60% fat diet,while the control group was given the control diet accordingly.After 10 weeks,Insulin resistance was observed to determine the molded case by measuring insulin resistance and lipid levels in blood.And then we will randomly divided the 10 molded rats,complying with the metabolic syndrome,into MS groups(n=5) and pioglitazone group(n= 5).MS group(n=5) was fed with 60% high-fat diet of Purification type +distilled water(20mg/kg,1/day);Meanwhile,Pioglitazone group(n=5)was given 60% high-fat diet of Purification type+pioglitazone(20mg/kg,1/ day);And the control group randomly choose 5 rats from NPD group(n=10),named control group n=5, continued with 60% fat diet-matched control diet+distilled water(20mg/kg, 1/day). During continuous feeding 4 weeks, Give each group of rats fed the corresponding gavage treatment at fixed time.After 14 weeks of feeding,We will detect rats blood glucose, insulin and blood lipid level. Take the visceral fat and blot moisture after weighing.And bservation of penile erectile function in rats by subcutaneous injection of apomorphine. And get the penis tissue to detect the total NOS activity and c GMP content.Observe the histologic changes.Results: The first 10 weeks,HFD group was significantly higher than NFD group in fasting plasma glucose、insulin and HOMA-IR index.There is a significant difference between the two groups(P<0.05). So we successfully established insulin resistance/hyperinsulinemia which is the core elements of the metabolic syndrome.At 14 weeks,the total NOS activity,c GMP content,APO-induced erection times and erection rate of MS group, were significantly lower than control group, there was statistical difference between the two groups(P<0.05).Meanwhile, MS group was significantly higher than control group in visceral fat weight, there were more significant differences between the two groups(P<0.05); 14 weeks, compared with MS group,PIO group was significantly lower in TC,which were significantly different compared with MS group(P<0.05) and the same thing happened in comparison between PIO group and control group;At 14 weeks,PIO group was significantly lower than MS group in insulin and HOMA-IR index.There is a significant difference between the two groups(P<0.05). but the total NOS activity,c GMP content,APO-induced erection times and erection frequency rate were no significant changes between the two groups,that showed no statistically significant difference(P> 0.05). Compared with control group, the cavernosum sinusoids, endothelial cells and smooth muscle cells of MS group and PIO group were lack of clear structure,the continuity of the vascular endothelial cell were damaged,and micro vascular were stenosis or occlusion.While PIO group’s cavernosum tissue blood sinus, endothelial cells and smooth muscle cell structure is not seen obviously improved compared with MS group,and the damagement of continuity of vascular endothelial cells, capillary lumen stenosis status has no obvious improvement.The data were analyzed by sigmastat 3.5 statistical software of English version,P<0.05 had statistical significance.Conclusion: Rats induced by high-fat diet may suffer from metabolic syndrome; And the metabolic syndrome led to rats appearing related performance of erectile dysfunction;A single application of pioglitazone in the treatment of metabolic syndrome can not improve the erectile dysfunction of syndrome metabolism source.
