Part one: The expression of TM4SF1 in breast cancer and its clinical significance Background and Objective:Breast cancer is the second common malignant tumor in the world, and is one of the most common malignant tumor in women,Seriously threat to the health of women.The new cases of breast cancer in 2012 alone is 1.67 million(about 25% of all cancer).Breast cancer is the fifth in all cancer deaths,but it is the most common cause of cancer deaths in women in the underdeveloped areas.In our country,the incidence of breast cancer has risen to the second of female malignant tumor in the city,and even risen to the first in some big cities.The incidence of disease is slightly lower than the city in the country.Breast cancer has become a big threat to women’s health.Along with the development of breast cancer more and more in-depth research, its development, metastasis and recurrence mechanism have been gradually known to mankind.TM4 SF family plays a different role in embryonic development, cell adhesion, migration, integration, cell activation and signal transduction process.In recent years,it has been reported about the TM4 SF family for development, invasion and metastasis of malignant tumor mechanism.Our study is intended to explore the expression of TM4SF1 gene in breast cancer tissue and the relationship with the histologic classification of breast cancer, the late shift. Methods1.The application of RT- PCR technique is to detect the expression of TM4SF1 gene in breast cancer, benign breast tumor and normal breast tissue.2.The immunohistochemical method is to detect the expression of TM4SF1 protein in breast cancer,benign breast tumor and normal tissue, and analyses whether there is a relationship about tm4sf1 family and history, age, lymph node metastasis, histopathological classification.3.Statistical analysis: SPSS17.0 statistical software is used to analysis the data we get. Results:1.The expression of TM4SF1 gene is significantly higher in breast cancer tissue than benign tumor and normal tissue; The expression is higher than normal tissue in benign tumor.2.TM4SF1 protein mainly expresses in cytoplasm and cell membrane also have a small amount of expression;The expression of tm4sf1 protein has nothing to do with age and lymph node metastasis;the expression rate in her-2 high expression type patients is significantly higher than other types of breast cancer. ConclusionTM4SF1 is high expression in breast cancer.Maybe it due to TM4SF1 is close relationship with the occurrence of breast cancer development and metastasis; It could promote the transfer of the tumor cells;The expression of tm4sf1 has nothing to do with age and lymph node metastasis,Maybe it is influenced by histopathologic types of breast cancer.Further studies are needed to prove our results;the expression rate of tm4sf1 in her-2 high expression type of patients is the highest in the types of breast cancer.may be there is a potential relationship between both, but still needs further exploring experiment.Part two: The expression of TM4SF1 in her-2 expression type of breast cancer and the analysis of related factors Background and Purposewe have got the conclusion that the expression rate of tm4sf1 in her-2 high expression type patients is significantly higher than other types of breast cancer from The first part of the experiment.We hypothesized that whether there is a relationship between TM4SF1 and her- 2 expression type of breast cancer?Whether TM4SF1 can influence the happen and development of her-2 expression type of breast cancer through some ways or some signaling pathways ?Whether TM4SF1 gene is able to influence clinical curative effect of Trastuzuma in treating the her-2 expression type of breast cancer.Therefore, we further explore the connection between Trastuzuma and tm4sf1 and the influence of related factors. Methods120 cases of HER- 2 expression type of breast cancer tissue paraffin section come from the pathology department of the first affiliated hospital of zhengzhou university,and Histopathology has been confirmed.Immunohistochemical method has been used to detect the expression of tm4sf1 protein in 120 cases paraffin section of breast cancer patients who are her- 2 high expression, used Trastuzuma.Operating procedure is strictly according to materials instructions.By immunohistochemical results evaluation standard we divided the results into two groups:high expression group(group A) and low/no expression group(group B).We Analyzed the difference of two groups in age, transfer rate, treatment condation, the clinical curative effect of Trastuzuma.Western blot method is used to detect the expression of tm4sf1 protein in different MDA-MB-453 lines with HER-2 high expression.this cell lines are disposed by different concentrations Trastuzuma. ResultsThe expression of TM4SF1 is related to the age and lymph node metastasis.the expression rate of tm4sf1 in breast cancer patients who are less than 50 years old is significantly higher than the patients with breast cancer who are more than 50 years old;the expression rate of tm4sf1 in breast cancer patients With lymph node metastasis is significantly higher than the patients without lymph node metastasis;TM4SF1 with curved bead sheet has no impact on the clinical curative effect of resistance.there is no difference about the expression of tm4sf1 gene and protein among different MDA-MB- 453 lines with HER-2 high expression which are disposed by different concentrations Trastuzuma. ConclusionTM4SF1 may program the infiltration and metastasis of breast cancer,and effect the prognosis of patients with breast cancer which is high expression of her-2 gene;Tm4sf1 does not affect the clinical curative effect of Trastuzumab,But it is expected to explore the treatment of breast cancer,Especially the new target of her-2 expression type of breast cancer.Provide a new treatment for inhibiting metastasis of breast cancer.
