Objective To investigate the effect of the different combination ofcompound Lizard Powder on Wnt2,beta-catenin,CyclinD1protein expression ofWnt pathway in rats with precancerous lesions of gastric carcinoma and to explorethe mechanism of the Compound lizards Powder in the treatment of PLGC.Methods120male SD rats were randomly divided into normal controlgroup, model group, the compound lizards powder (including80mesh,100meshand80+100mesh mixing equal paste suspension) group and Vitacoenzymetreatment group of20. Except for the normal control group, MNNG and othercomprehensive factors(including irregular diet and emotional stimulation) wereused to establish precancerous lesions of gastric carcinoma(PLGC) model,8weeks.After the rat model was established,80mesh,100mesh,80mesh and100mesh mixture of the compound lizards powder, normal saline and vitacoenzymewere applied on the PLGC model, respectively,12weeks. The protein expression ofWnt2,beta-catenin and CyclinD1was detected using immunohistochemical method.The pathological and histological changes of gastric mucosa were observed withoptical microscope.Results The optical density of immunohistochemical image was calculatedthrough the analysis of the Image-the Image Pro Plus6.0system. The expermentalresults showed that:the positive expression of cell proliferation protein Wnt2,beta-catenin and CyclinD1in model group was significantly higher than that the normal group(P<0.01).The positive expression of cell proliferation proteinWnt2,beta-catenin and CyclinD1protein in each group treated with the compoundlizards powder and vitacoenzyme group was significantly lower than that of themodel group (P<0.05). The positive expression of the three kinds of proteins in thedifferent combination of compound lizard powder(DCCLP) was significantlylower than the compound lizards powder (TCLP)80mesh group and TCLP100mesh group (P<0.05).Conclusions (1) DCCLP could protect and repair the damaged gastricmucosa,improve or partly reverse the pathological changes of gastric mucosatissue morphology.(2)DCCLP could effectively reduce or inhibit the proliferationprotein expression level of Wnt2,beta-catenin,CyclinD1.(3)This revealed thatDCCLP could repair the damaged of gastric mucosa and treat or reverse thedevelopment of PLGC, of which the mechanism might be that the excessiveproliferation of Wnt2,beta-catenin and CyclinD1was inhibited through theintervention of the proliferation and differentiation of the mucosa to promote cellapoptosis and restore the balance of cell proliferation and apoptosis, thus blockthe precancerous lesions of stomach cancer.(4)DCCLP curatived effect on thedamaged gastric mucosa was better than the compound lizards powder80meshparticle agent and100mesh particle agent, that traditional Chinese medicinecompound preparations could improve the curative effect of particles through theform of combination.
