Objective: To assess the genotypic distribution of single nucleotide polymorphisms(SNPs) in CFH and HTRA1in the population of Ningxia, and whether the interactionbetween age-related macular degeneration susceptibility, as well as genetic andenvironmental factors.Methods: For age, gender and ethnic matched case-control association studies,select tag SNPs sites:CFH gene(rs800292and rs1410996),HTRA1gene (rs11200638).SNP genotyping of150cases of AMD patients and150normal controls were determinedby Sequenom mass spectrometry analysis platform to explore the relationships betweenSNPs and AMD in Ningxia population.The environmental factors for AMD and theinteraction between environmental factors and the two genes were analyzed bymultivariate unconditional logistic regression.Results: The risk of point C allele frequency was74.1%,59.7%and67.8%,54.1%in the case group and control group CFH rs800292and rs1410996.The a risk allelefrequency for HTRA1rs11200638in case and control groups were68.6%and46%respectively,which were significantly higher than that in the control group,And thedifferences between case and control groups were were significantly.The CFH andHTRA1gene has significant difference to the patients with wet AMD. Multivariatelogistic regression analysis found that the five variables which were age, ethnicity,gender, smoking, high blood pressure eventually into the model.A higher proportion ofHan in the pathogenesis of AMD.Logistic regression analysis: there is no interaction(P>0.05) between genes and they are independent dangerous effects. Analysis the interaction between genes and environment indicated that: adjust the other environmentalfactors and genotype, Various environmental factors and rs11200638, rs800292,rs1410996does not exist interactions. The rs800292and rs1410996for CFH gene havelow degree of linkage disequilibrium.Conclusion:1.The AMD in Ningxia populations associate with the polymorphism ofrs800292,rs1410996for CFH and rs11200638for HTRA1.2. Each gene-gene, gene-environmental factors there is no interaction. |
