Objective: To establish the accelerative and early type Ⅱ diabeticnephropathy model and observe the protective effect and mechanism ofapocynin on glomerular of early type Ⅱ diabetic nephropathy mice.Methods:1, Perform the unilateral nephrectomy surgery on8weeks olddb/db mice in order to establish the accelerative and early type Ⅱ diabeticnephropathy model.2, Induced type Ⅱ diabetic nephropathy mice wereassigned into3groups, vehicle, apocynin and positive control pioglitazonefor8weeks treatment respectively. Measure6hrs fasting blood glucoseand body weight every2weeks and24hrs albuminuria every4weeksduring the treatment period. Measure glomerular filtration rate and observepathological changes of glomeruli in order to determinate the protectiveeffect of apocynin on kidney after the last administration. Results:1, Positive control pioglitazone group showed an effect on reducing bloodglucose and lower blood HbA1C level after8weeks treatment comparedwith vehicle group (P<0.01); There is significant differences in24hrs albuminuria between positive control pioglitazone group after8weekstreatment compared with vehicle group (P<0.01); Glomerular filtration rateof positive control pioglitazone group maintained at the normal level butvehicle group decreased after8weeks treatment; Positivecontrol pioglitazone group showed an effect on reducing pathologicalchanges of glomeruli after8weeks treatment compared with vehiclegroup.2,Pioglitazone and apocynin treatment group showed a stable trendon blood glucose and there are differences compared with vehiclegroup (P<0.05); There is significant differences in24hrs albuminuria between apocynin group after8weeks treatment compared with vehiclegroup (P<0.01); Glomerular filtration rate of apocynin group maintained atthe normal level; The results of renal PAS staining: the vehicle controlgroup showed the obvious glomerular sclerosis and mesangial proliferation.Apocynin group also glomerular pathology significantly reduced.Conclusion: apocynin can effectively maintain stable blood glucose level,reduce the24hrs albuminuria and prevent glomerular produces thecorresponding lesions.
