| 2-HPA is a kind of monoamine oxidase B inhibitors, a new drug of medicineclassification class1. It’s use for Parkinson’s disease, muscular atrophy of amyotrophiclateral sclerosis. It can protect nerve of cerebral apoplexy on animal models.2-HA was propargylated to get2–HPA. After make2-HPA into salt, we use GS-MSto determine the composition of the synthetic material. SD rats were divided into controlgroup, low dose group, high dose group. We gave rats intraperitoneal injection of0.9%saline,10mg/kg2-HPA,2-HPA20mg/kg respectively. Experiments were divided intoshort-term and long-term experiment.Short-term experiment continued7days. Six days for drugs, we test the ability of newobjects on7th day. After animals were handled, we exam dendritic spines of Golgi, examnewborn neurons of immunohistochemistry. Explore the effect of2-HPA on protein levelsto investigate whether affect the related functions of learning and memory.Long-term experiment continued28days. We test the ability of new objects on23thday. Morris water maze behavior method observed the change of learning and memoryability from24th to28th day. Another group were tested on eight arm maze to inspect worklearning and memory of rats. After28days, rats were handled for testing the changes ofdendritic spines, newborn neurons, BDNF protein level, NMDAR2A, NMDAR2B,p-CREB/CREB to investigate whether2-HPA affect the related functions of learning andmemory.The result shows that2-HPA did not show obvious ability to improve learning andmemory in short-term experiment. But in long-term experiment2-HPA showed obviousability to improve learning and memory. To Further explore the neural morphology changesin neural morphology experiment, using Golgi method to investigate the change of thenumber of dendritic spines, low dose group and high dose group are significantly higherthan the control group. In immunohistochemistry experiment, newborn neurons number oflow dose group and high dose group are significantly higher than the control group. Tofurther explore the physiological mechanism, we used Western Blot to detect BDNF,NMDAR2A, NMDAR2B, p-CREB/CREB. BDNF, NMDAR2A, p-CREB/CREB levelsof low dose group and high dose group were obviously higher than control group. 2-HPA have can raise the level of BDNF, NMDAR2A, and CREB phosphorylationlevel to affect neural plasticity, neurogenesis. So the ability of work learning and memorywas improved. |
