The asthma experiments of animal are an important experimental step of drug research forasthma, it can reflect the asthma symptoms and evaluate the drugs of asthma effectively. Thetraditional measurement method is done in anesthesia animals, the measurements can notreflect the physiological states of the animal, while the conscious animal model can overcomethis shortcoming. In this paper, we established an asthma model of conscious puppies for drugevaluation, and compare the reaction of conscious puppies and anesthetized puppies in asthmaexperiment. The content of research is as follows:(1) We select puppies as animal models, andselect the ITS wireless telemetry system.Make sure that the surgical instruments is sterile before the operation, and animal can getenough time and reasonable diet for rest, guarantee the animals cure to their normal physicalcondition, attention in the process of surgery after the surgery to give animals to get enoughtime and reasonable diet.(2) We design an asthma system of conscious puppies, including the tidal volume (head-outplethysmograph, moisture measurement module, airway flow measurement module,multifunctional physiological parameters device and atomization dosing module).The devicecan directly detect respiratory function indicators, such as airway flow, tidal volume and intrapleural pressure. The airway resistance and lung compliance can be obtained by computing.The system can reflect the asthma reactions in puppies effectively.(3) The hypoxia experiments have been performed in conscious puppies. The mixed gas (4%CO2,21%O2and75%N2) has been ventilated for10minutes, puppies are in anoxic condition,and the physiological change in hypoxia state has been recorded. Then the normal air hasbeen ventilated, the physiological change in recovery state has been recorded. In hypoxia state,the respiratory rate increase43%in10minutes, the heart rate fell by13%, intrathoracicpressure increase75%, the airway flow increase87%and the tidal volume increase66%in10minutes. When giving animal normal air, the parameters back to normal in nearly3minutes.(4) The histamine induce asthma experiment are performed in conscious puppies andanesthesia puppies. The result show that the airway resistance increased152%when injectedhistamine. When injected histamine after inhaled ventolin, the airway resistance only increase 18%.Anesthesia experiment use the same dosage of histamine, the airway resistance ofanimals increased67%. When injected histamine after inhaled ventolin, airway resistanceincreased20%.These proved that the system can also be useful for pulmonary function testingin anesthesia animals. Compared with the data between the conscious state and anesthesiastate, we found that after administration of histamine the increment of airway resistance inconscious states more obvious than in anesthesia state, the asthma reaction of consciousnessanimals is more serious than anesthesia.The experiment of consciousness animals is more accord with the reaction of animal, theestablishment of a system of conscious puppies for drug evaluation has more importantsignificance of research.
