KHB is a commonly used traditional Chinese medicine and possesses thefunctions of antipyretic, detumescence, analgesia and anti-inflammatory and so on. Inrecent years, with research of KHB, its much chemical components andpharmacological activities has been known, but its water soluble constituents andpharmacological activity remains to be known little. This dissertation topic lucubratesin water extract-alcohol precipitation part chemical composition of KHB and itsactivities, laid the groundwork for new drug development. The dissertation includesthree parts:First, we have made systematically inspection and identification work on thechemical composition of water extract-alcohol precipitation part of KHB anddetermind which contains much polysaccharides, flavonoids ingredients, littleinorganic salt and amino acid, and the content of polysaccharides is about20%, thecontent of flavonoids is about9%, the content of amino acids is about2%and thecontent of inorganic elements is about6.8%.Second, using TOYOPEARL HW–40C gel filtration chromatography, the waterextract-alcohol precipitation part was divided into KHB-1、KHB-2、KHB-3parts, andKHB-1parts divided into KHB-A、KHB-B、KHB-C、KHB-D、KHB-E parts withDEAE-52ion exchange chromatography. The biological activity of all parts werescreened by BIAcore, the result shows the active ingredient concentrate in partKHB-1, KHB-C、KHB-D、KHB-E parts have higher activity, especially activity ofKHB-E increased7.4times than water extract-alcohol precipitation parts. In theprocess of separation and purification, we have obtained a compound in part KHB-2which is identified as sodium oxalate.Third, polysaccharide as index, effective components extraction and purificationprocess was optimized. The extraction process was optimized by orthogonalexperiment, compared the macroporous adsorption resin and alcohol precipitationpurification process, determined the optimum extraction and purification process forKHB, with15times amount of water, under60℃, extract90min, extractconcentrate to relative density1.16-1.18, make the alcohol content reach85%,sedimentate8to10hours, leaching precipitation, volatilize ethanol to prepareeffective parts.
