Objective To observe the effects of Eldepryl on expression of inflammatory factorTNF-α、IL-1βand IFN-γin nigra of Parkinson’s disease model rats, to explore theparking pathogenesis of eldepryl for Parkinson’s disease in treatment.Methods1A total of96SD rats were randomly divided into control group, Parkinson’sdisease model group, Levodopa treatment group and Eldepryl treatmentgroup. Eachgroup was divided into4days and8days subgroups after the success of modelpreparation(n=6).2Parkinson’s disease rat model were established by injectingrotenone in subcutaneous. Dissolved the rotenone in the sunflower oil, made it mixedfully, then configured2mg/ml rotenone sunflower oil emulsion and injectied it in thesubcutaneous tissue of each rat daily by2mg/kg. Ready to observe and record thebehavior changes after the rote-none was given, we terminated the drug once occurredthe behavioral scoring as soon as possible. Carried on the behavior study examinationaccording to the Parkinson animal model behavior study grading standard.3Controlgroup and PD group: injected the sunflower oil (2mg/kg) in hypodermic tissecontinuously, after a successful stay model preparation, stopped subcutaneous and filledthe physiological saline into the stomach. Levodopa treatment group: fill the levodopa(10mg/kg) into the stomach each rat. Eldepryl treatment group: fill the eldepryl (0.5mg/kg) into the stomach each rat.4We detected the expression of TNF-α, IL-1β andIFN-γ in substantia nigra in each group by Immunohistochemistry and Western blotting.5We observe tissue slices with microscope camera OLYMPUS (400×), then analysisthem by Motic Med6.0digital medical image analysis system. The results of WesternBlot were analyzed with Image J.6Application SPSS17.0statistical software for dataanalysis at last. Measurement data are presented as mean±SD(x s), according to thedata for normality Wtest and homogeneity of variance Levene test, using randomizedblock design analysis of varianc, P<0.05for the difference was statistically significant.Results1The expression of TNF-α, IL-1βand IFN-γin nigra were increased in theParkinson’s disease model group compared with that in the control group (P<0.01), andthere were more expression of TNF-α, IL-1βand IFN-γat8d compared with those at4d (all P>0.05).2The expression of TNF-α, IL-1βand IFN-γin nigra were reducedin the Eldepryl treatment group and the Levodopa treatment group compared with that in the Parkinson’s disease model group (P<0.05), and there were less expression of TNF-α,IL-1βand IFN-γat8d compared with those at4d (all P<0.05).3The expression ofTNF-α, IL-1βand IFN-γin nigra were reduced in the Eldepryl treatment groupcompared with that in the Levodopa treatment group (P<0.05), and there were lessexpression of TNF-α, IL-1βand IFN-γat8d compared with those at4d (all P<0.05).Conclusions1TNF-α, IL-1βand IFN-γin nigra were increased in the Parkinson’sdisease model group in nigra, inflammatory factor may be related to Parkinson’s disease.2By reducing the expression of TNF-α, IL-1βand IFN-γin nigra in rat model of PD,Eldepryl could treat Parkinson’s disease.
