Objective:To study the dynamic changes of the cytokines IL-17,IFN-γ,IL-2in related HLA-haploidentical non T cell-depleted in vitro peripheral hematopoietic stem cell transplantation (RHNT-PSCT), and analyze they where or not associated relevant to acute graft-versus-host disease (aGVHD).Method:To check the cytokines (IL-17,IFN-γ,IL-2) level of21patients who underwent the RHNT-PSCT we designed in the patients serum, by enzyme-linked immunosorbentassay(ELISA) at different time points, and compared with that of the22patients who taked the HLA-matched sibling non T cell-depleted in vitro peripheral blood hematopoietic stem cell transplantion (MSNT-PSCT) simultaneously. Result:1. In related HLA haploidentical(RH) patients without aGVHD, the concentration of IL-2after transplant was no significant changes than that in pre-transplant, the concentration of IFN-y increased at30and60days, but recovered at90days compared with that in pre-transplant, while the concentration of IL-17was higher than that in pre-transplant at90day(P<0.05); in HLA-matched silbing (MS) patients without aGVHD, the concentration of IL-2after transplant increased than that in pre-transplant(P<0.05), while the concentration of IFN-y and IL-17had no significant change.2.1n RH patients with aGVHD, the concentration of IL-2and IL-17after transplant increased significantly than that in pre-transplant(P<0.05), and IFN-y decreased than that in pre-transplant (P<0.05); in MS patients with aGVHD, the concentration of IL-2and IFN-y after transplant increased significantly than that in pre-transplant(P<0.05), while the concentration of IL-17had no significant changes than that in pre-transplant. Conclusion:Compared to MSNT-PSCT, the changes of IL-17, IFN-y and IL-2in our RHNT-PSCT with or without aGVHD is different partially.
