Objective:This study aimed to compare the expression and distribution fo TGF-β1,P-Smad2,3, Smad-7and types Ⅰ and Ⅲ collagen protein in the peripheral and central areas of human skin hypertrophic scar, and analyze the biological significance in order to guide the application of clinical. Methods:A total of20cases with human skin HS admitted at the Departmant of Plastic Surgery, and all skin tissues divide into three groups, include central and peripheral areas and nomal. Then the expression of TGF-β1,P-Smad2,3, Smad-7and types Ⅰ and Ⅲ collagen protein was detected with western blot method. Results:Compared with normal skin groups (2.47±0.23), the expression of TGF-β1protein in central groups and peripheral groups is (2.52±0.22)、(3.83±0.35).There is significant difference between two groups (P=0.001). Compared with normal skin groups (5.01±0.51), the expression of Smad7protein in central groups and peripheral groups is(3.16±0.31)、(1.87±0.19).There is significant difference between two groups (P=0). Compared with normal skin groups (0.75±0.07), the expression of P-Smad2protein in central groups and peripheral groups is (1.43±0.09)(2.04±0.24).There is significant difference between two groups (P=0). Compared with normal skin groups (2.47±0.33), the expression of P-Smad3protein in central groups and peripheral groups is (2.4±0.29)、(3.72±0.36).There is significant difference between two groups (P=0.04).Compared with normal skin groups (1.24±0.13), the expression of types Ⅰ collagen protein in central groups and peripheral groups is (1.99±0.12)(1.80±0.17).There is significant difference between two groups (P=0.02). Compared with normal skin groups (1.83±0.16), the expression of types Ⅲ collagen protein in central groups and peripheral groups is (2.43±0.40)、(2.34±0.38).There is no significant difference between two groups (P=0.137). Conclusion:Compared with the peripheral and central area of HS, the TGF-β1and P-Smad2,3protein expression much more higher, but the Smad7protein expression much more lower, the type Ⅰ and Ⅲ collagen expression is no significant difference between two groups. In the clinical, we can focus on the therapy of peripheral area may lead to better treatment.
