Objective:Research the gender differences in different valence state arsenic infected rats brain tissue of arsenic metabolites and arsenic metabolism related enzymes, in order to further clarify the arsenic metabolism process and toxic mechanism lay the foundation.Methods:The70healthy7-week-old Wistar rats of clean grade were randomly divided into7groups, namely the control (deionized water) group and20,6.7,2.2mg/kg sodium arsenite (iAs3+) and sodium arsenite (iAs5+) exposed groups of10, male and female. Way of using the free water exposure, continuous exposure three months after death, blood samples were collected to measure serum oxidized glutathione (GSSG) using enzyme-linked immunosorbent assay (ELISA), reduced glutathione (GSH), glutathione peroxidase (GSH-PX), glutathione S-transferase (GST), glycolytic enzymes (GE) and methyl transferase (As3MT) glutathione reductase (GR) changes, using high performance liquid chromatography-hydride generation atomic fluorescence spectrometry (HPLC-HGAFS) Determination of arsenic species in brain tissue product method [iAs3+, iAs5+, monomethyl arsenic acid (MMA), dimethyl arsenate (DMA) and total arsenic (TAs)] content.Results:l.Sodium arsenite high dose group of the second month, in the first three months of high-dose and high-dose group arsenite the first three months of weight gain in male rats is higher than females, the difference was significant(P<0.05).2.Sodium arsenite low and high dose group and arsenite exposure groups of male rats blood iAs3+were higher than female rats, the difference was statistically significant(P<0.05).3.Each dose of sodium arsenite, sodium arsenate in the low dose group of male rats DMA blood, TAs content and sodium arsenite dose Asia, arsenic, sodium in the blood of male rats exposed groups were higher than those of female MMA, arsenic sodium, and high dose groups of male rats in the DMA blood, TAs content and low sodium arsenite dose group of male rats lower than female MMA content in the blood, the difference was significant (P<0.05).4.Control group, sodium arsenite and sodium arsenite low dose, medium dose group of male rat brain tissue iAs3+levels were lower than in female rats, high doses of sodium arsenite and sodium arsenite exposure groups of male iAs3+rat brain tissue were higher than female rats, the difference was significant(P<0.05).5.Low High doses of sodium arsenite, sodium arsenite high dose group of male rat brain tissue content and sodium arsenite DMA high doses, arsenite dose group in male rat brain tissue TAs content below female rats, arsenite low dose group of male rat brain tissue DMA content and low doses of sodium arsenite, arsenic and sodium content of the high dose group and MMA arsenite low-dose male rat brain tissue brain tissue of male rats exposed groups of organic arsenic levels higher than female rats, the difference was significant(P<0.05).6.Control group, high dose of sodium arsenite, sodium arsenite low dose group of male rats in blood GSSG content of less than female rats, low doses of arsenic, sodium in the blood of male rats exposed groups were higher than females PX the difference was significant(P<0.05).7.Sodium arsenite low dose group of male rat brain tissue As3MT vitality dose brain tissue GSH content is lower than female rats in the control group, arsenite rat brain tissue in the high dose group and GSSG levels control group, low-dose sodium arsenite exposure group, arsenic sodium dose group of male rat brain tissue GST was higher than female rats the difference was significant(.P<0.05).Conclusion:Increases with increasing arsenic product content in the blood exposure to arsenic, and through the blood brain barrier in the brain tissue accumulation of arsenic metabolites content increased. Susceptibility to arsenic in male rats with stronger, more easy to accumulate in the body than in female rats, and a strong inorganic arsenic methylation in male rats in female rats, there is a gender difference in the female, male rats under the same exposure to arsenic tips, and arsenic metabolism in male rats cytotoxicity than females.
