Objective:This study is to find out the endogenous mechanism of histoneacetylase p300ã€CBP by moxibustion in myocardial apoptosis after heavy exercise, and will be used to protect the heart during heavy sports training, by observing the apoptosis-related factors Fas, Bcl-2and p300ã€CBP histone deacetylase apoptosis relationship to explore p300ã€CBP histone deacetylase anti-apoptotic role in the myocardium moxibustion, as in sports training to effectively protect the heart provides a new research ideas.Methods:60adult KM mice were randomly divided into blank control group (A), exercise control group (B), exercise and moxibustion therapy group (C)(n=20).Both group B and group C kept swimming training every day, while group A was free activity during the time.In the first3days, let the mice swim for30minutes a day adaptation.24h after the last swimming training,the three groups of mice were killed off the vertebral quickly take heart tissue, paraffin embedded, sliced for immunohistochemical detection of Fas, Bcl-2, p300, CBP expression,, and cardiomyocyte apoptosis detection.Results:(1)Fas protein expression:B Compared with A, the Fas protein expression was significantly increased (P<0.05); C compared with B, the Fas protein expression was significantly decreased (P<0.05), C Compared with A, the result is insignificant(P>0.05).(2). Bcl-2protein expression:B Compared with the A, the Bcl-2protein expression was significantly decreased (P<0.05); Bcl-2protein expression increased significantly when C compared with B (P<0.05).(3)p300expression:A Compared with C, p300expression was significantly decreased (P<0.01); p300expression of C was significantly higher than B (P<0.05).(4) CBP expression:B Compared with A, CBP expression was significantly decreased (P<0.05); C was significantly higher than B (P<0.05).(5) Apoptosis Detection:B Compared with A, apoptosis rate was significantly increased (P<0.01);there is significance between A and C(P<0.05); apoptosis rate of C was significantly decreased than B (P<0.05).Conclusion:Moxibustion can promote the expression of p300ã€CBP in mouse myocardial cells after heavy training, thereby inhibiting the apoptotic process started, so that decreased expression of death receptor Fas, increased expression of Bcl-2,so that to decreased the myocardial cells apoptosis rate after training and protect heart function. |
