Calpains is a high regulative Ca2+-dependent proteases system that will promote muscular fibril disaggregation when hydrolization. In calpain system, calpain activator is the positive regulator and it can play a important role in activate calpain. Research indicated that UK114, the goat liver tumour antigen,is the activator of calpain-1. UK114, a14.5kD protein, is a member of a family of conserved low-molecular-mass proteins, and it’s widely distributed in prokaryotes and eukaryotes.These proteins that are usually composed of130amino acids, has highly expressed in liver and kidney, in comparison, has lowly expressed in cancer tissues of the liver cells. It is significantly down-regulated in tumor tissue of hepatocellular carcinoma as compared to the adjacent normal tissue. Our research group has done some research early This research investigated the anti-tumor effect of UK114through vitro and vivo experiments.We observe the effect of UK114on Ascites tumor mice inhibition and the life prolongation respectively. the UK114was proved the positive effect on anti-tumor, We saw the phenomenon of tumor cell apoptosis.In this experiment,we choosed Hela cells and normal liver cells of Human.RNA interference、real-time PCR%EGTA treatment、cell coun、plate clone formation were performed to measure the effect of UK114on calpain We found that the expression reducing of UK114or calpain were both hindered the growth of Tumor cell and the ability of plate clone formation...
