| Background:Intrahepatic cholestasis is a clinical common disease,frequently-occurring disease, its formation mechanism is very complex, including low expression and dysfunction of liver cell membrane transport protein,oxidative stress,inflammatory lesions,liver cells structural abnormalities,and many other aspects.Among them,the dysfunction of liver cell membrane transport protein expression and is the main pathogenesis,as other secondary pathogenesis.At present studies have found that liver cells in the basement membrane of the two kinds of bile composition intake system,including NTCP, BSEP,play an important role in the formation of intrahepatic cholestasis.At the same time a large number of clinical studies have shown that large dose of chishao can relieve intrahepatic cholestasis.This study by observing the different doses of chishao in alpha isothiocyanate of naphthalene ester (ANIT) induced intrahepatic cholestasis in the rat model of liver cell membrane transporters NTCP,the influence of BSEP,discusses its treatment effect and potential mechanism of action.Research objective:To observe differentence of chishao,ANIT induced intrahepatic cholestasis rats model of the improvement of liver tissue pathology,and the effect on liver cell membrane transporters NTCP, BSEP.Research Methods:The30male SD rats,and randomly divided into normal group, model group, model+low chishao, model+middle chishao,model+high chishao, each group has5rats only. ANIT (5ml/kg) of disposable lavage in the rat model with acute intrahepatic cholestasis.Respectively to gastric drug intervention, the specific dosage:low chishao group (10g/kg), middle chishao group (20g/kg), high chishao group (40g/kg), UDCA group (40g/kg). Drug intervention after7days, generally, liver tissue pathology, and liver of rats NTCP, BSEP protein and mRNA expression.Research results:The SD rats to ANIT disposable lavage can be successfully established model for intrahepatic cholestasis. After drug intervention, radix paeoniae rubra and UDCA group liver tissue pathological damage, which the pathology to improve the degree of radix paeoniae rubra group related to the dose.Liver NTCP, BSEP expression radix paeoniae rubra group were higher than model group, among them, the NTCP expression positively related to the dose, the expression of BSEP dose not significantly related.Research conclusions:Chishao decoction can increase ANIT NTCP induced intrahepatic cholestasis model of liver cells, the expression of BSEP, relieve the damage of liver tissue pathology NTCP expression positively related to the dose, the expression of BSEP dose not significantly related. |
