Background and Significance:The bone defect remains as a clinical challenge that facing multiple operations, insufficient autologous bone graft, and long-term use of antibiotics. To develop a new biomaterial with the bone repairing ability would be helpful for the management of bone defect.Objective:To evaluate the topographical cell guidance of oriented silk fibroin nanofibers on BMSC proliferation and the action of the cytoskeleton.Methods:Oriented and random-aligned silk fibroin fibrous scaffolds can both be obtained via electrospin with rotating collectors. BMSCs were then cultured on the surface of this material. IHC and SEM observed cell morphology and structures of the cytoskeleton. With MTT assay, the influence of topography on cell proliferation was also investigated.Results:The tensile experiments showed mechanically anistrophic features of the oriented silk fibroin nanofibers. BMSCs, which were cultured for3days on oriented fibrous scaffolds, were aligned according to the long axis of silk fibroin fibers on the surface and compared to non-aligned scaffolds. It was also observed that BMSCs cultured on oriented scaffolds had a higher rate of proliferation, which showed to be statistically significant after3days compared with the silk films or after4days in both comparisons.Conclusion:Electrospining oriented silk fibroin scaffolds were capable of inducing BMSCs alignment and promoting BMSCs proliferation.
