Objective:To observe the effect on protein expression of SDF-1and CXCR4in ischemic cerebral infarction model rats treated by Icariin,and to explore the mechanisms of protecting ischemic brain tissue by SDF-1and CXCR4.Methods:Using the method of plugging wire into SD rats’ middle cerebral artery to make cerebral infarction model which as experimental object.Those SD rats were divided into5groups including the sham-operated group,the model group,the low dosage group,the middle dosage group and the high dosage group which has12SD rats in each group.each group was randomly divided into three time points(n=4):3d,7d,14d.The sham-operated and model group were administrated physiological saline by stomach,and the experimental group were administrated Icariin by stomach.Then the changes of neurobehavioral scores were observed in3d,7d,14d.And choose the best one to observe the infarct size and the protein expression of SDF-1and CXCR4in cerebral infarction model rats which were detected by the western-blot method.Results:(1)Compared with the model group,and the effect on neurobehavioral score of the low dosage group are more better(P<0.05).(2)Compared with the model group,the infarct size of the low dosage group are more less(P<0.05).(3)The protein expression of SDF-1and CXCR4in model rats’ brain tissue are based on level expression.In the low dosage group rats’ brain tissue, the SDF-1and CXCR4expression are higher than model group(P<0.05).Conclusion:(1)There has more significantly effect to improve cerebral infarction model rats neurological damage by low dosage Icariin.(2)There has more significantly effect to reduce the infarct size by low dosage Icariin.(3)The low dosage Icariin group can significantly increase the protein expression of SDF-1and CXCR4in brain tissue.(4)Those maybe the mechanisms of neuroprotection of cerebral infarction by low dosage Icariin to increase protein expression of SDF-1and CXCR4in ischemic tissue.
