Objective:To explore the curative effect of glucocorticoid (GC) in thetreatment of Duchenne muscular dystrophy (DMD) and its possiblemechanism effectively.Methods: Mdx mice as the research object, were divided into the GCtreatment group and the placebo group by the method of randomized、controlled, then observe the changes of muscle strength after GC treatmentthrough suspension test, CK value understanding muscle damage change,HE staining and electron microscope observe the changes of muscle tissuepathological;Using Western blotting utrophin detect muscle tissue proteinexpression level changes and the nf-kappa B/I-kappa B predominate, therelationship between HSP70protein expression level changes.Results:Compared with the placebo group:(1) The muscle strengthand CK values of GC treated mdx mice were improved (P <0.05);(2)Themuscle histopathologic of mdx mice have improved after GC treatment;(3)The expression levels of HSP70protein and Utrophin protein mdx mice muscle were increased after GC treatment (P <0.05), there is a decrease inthe expression levels nf-kappa B protein after GC treatment (P <0.05);whilecompared the expression levels of I-kappa B protein with untreated groupin the muscle tissue, there is no statistical difference.Conclusion: GC treatment can improve the muscle strength of mdxmice;GC treatment Can increase the expression of HSP70,reduce theactivity of nf-kappa B, in order to promote utrophin expression, thus reducemuscle lesions.
