Objective: To research the effect of COX-2on Acsl family related gene expressionand confirm the regulated targets of COX-2on lipid metabolism of the HSC-T6cells.Methods: pYr-1.1-COX2-shRNA plasmid and pYr-1.1-NC vector were stabilytransfected into HSC-T6cells by liofectamine. Fluorescent inverted microscope wasused to observe the transfection efficiency at48h after transfection. The expression ofCOX-2, Acsl1,3,4,5and6were detected by Real time PCR, and select the significantgenes in Acsl family according to the Real-time PCR results and the proteinexpression of Acsl in HSC-T6by Western Blot.Results: The green fluorescence can be observed in HSC-T6cells cytoplasm at48hafter transfection and the transfection efficiency is more than75%. Compared to mockgroup and negative control group, the mRNA levels of COX-2inpYr-1.1-COX2-shRNA group were significantly reduced at48h aftertransfection(P<0.05). The mRNA and protein levels of Long-chain fatty acyl-CoAsynthetases6(Acsl6) were significantly increased in pYr-1.1-COX2-shRNAgroup(p<0.05), while the expression of Acsl1,3,4,5have no significantly change(p>0.05).Conclusion: Knockdown of COX-2can significantly increased the expression ofAcsl6, which may involved the regulation of lipid metabolism in hepatic stellate cells.
