Objectives1. Explore the polymorphisms of KCNH2gene’s influencing to acquired atrialfibrillation, provide the evidence for gene therapy of atrial fibrillation.2. Provide frequency data of SNP of KCNH2for the Hunan region, which alsoprovide clinical information and date for future epidemiological studies of atrialfibrillation.Materials and MethodsCase control study is based on hospital cases, matching1:1, select KCNH2rs1805120by genome-wide association studies, and have correlation analysis on it.The above cases are from the First Affiliated Hospital of Nanhua University fromJune1,2012to December30,2012. The diagnostic criteria of AF: base on ECG, andhave a history of atrial fibrillation. Control group are selected from the same wardwith atrial fibrillation patients without a history of atrial fibrillation andelectrocardiogram without AF performers. Finally, we collect about98in the AFgroup,102cases in the control group. Matching conditions: the same sex, age andthe fundamental heart disease is homogeneous. Get blood samples, use ACDanticoagulant, extract DNA, and then applicate Wuhan gene’s technology platformfor selecting SNP genotyping and determining their genetic model, using Logisticregression analysis SNP sites and other factors influence on atrial fibrillation.ResultsIn the atrial fibrillation groups: the frequency of allele C is accounted for33.7% (66cases)while the allele T is accounted for (66.3%)(130cases); The control group:the frequency of allele C is accounted for29.4%(60cases), while the allele T isaccounted for70.1%(144cases), the difference of allele frequency between the twogroups has no statistical significance. Genotype frequency distribution statistics: Inatrial fibrillation group, CC genotype frequencies is accounted for10.2%(10cases),C/T genotype frequencies is accounted for46.9%(46cases), TT genotype frequenciesis accounted for42.9%(42cases); The control group: CC genotype frequencies isaccounted for7.8%(8cases), C/T genotype frequencies is accounted for45.1%(46cases), TT genotype frequencies is accounted for47.1%(48cases), by the χ2squaredtest analysis, genotype frequencies distribution between the two groups has nostatistical significance.Conclusions1. The polymorphisms of KCNH2gene rs1805120may be no correlation withatrial fibrillation in Hengyang.2. Left atrial diameter may increase the incidence of atrial fibrillation, Theenlargement left atrial diameter may be a risk factor for atrial fibrillation...
