Objective: To study the relationship between the brain edema, the expression ofAQP-4and the damage degree of the blood-brain barrier on the middle third of thesuperior sagittal sinus and its reflux veins occlusion in rats; and explore whethersodium aescinate can reduce brain edema through regulating the expression of AQP-4in brain tissue after sinus.Methods:96healthy male SD rats were divided randomly into four groups:(1)Sham Operation(SO)group(n=6);(2) the middle third of the superior sagittal sinusand its reflux veins occlusion (SSS+RV/SR) group(n=30);(3) Normal saline treatedto the middle third of the superior sagittal sinus and its reflux veins occlusion(NS+SSS+RV/NS) group(n=30);(4) Sodium aescinate treated to the middle third ofthe superior sagittal sinus and its reflux veins occlusion (SA+SSS+RV/SA)group(n=30). At the same time, as to the group SR, the group NS, the group SA, wedivided each group again into five subgroups according to preconcertedtime:6h(n=6),12h(n=6),24h(n=6),48h(n=6),72h(n=6). The neurological functionsof rats were observed before the rats were killed,the histological changes ofparasagittal brain tissue were evaluated in experimental animals,the water content inbrain tissues were measured by wet and dry weight method,detected the contentof EB of parasagittal brain tissue what can reflect the blood-brain barrierpermeability. Immunohistochemistry was used to determine the change ofexpression of AQP-4protein.Results:1) Rats of group SO showed no neurological deficit, while group SRand group NS were begin appeared neurological disorders after6hours in themiddle third of the superior sagittal sinus and its reflux veins occlusion, and it hadtime-dependently increased. At24hours, the neurological function score get highest,decreased gradually after48hours, and at72hours, the neurological function score significantly lower than24hours. As to each preconcerted time subgroup of thegroup SA, neurological deficit is lighter than SR and group NS (P<0.05);2) As to the rat of SR group and group NS, after6hours in the middle third ofthe superior sagittal sinus and its reflux veins occlusion, HE staining showed thatneuron edema, degeneration, necrosis, and Nissl staining showed that the number ofNissl bodies decreased, both were get the most obvious at24hours. Then, neurondegeneration get better than before, and the number of Nissl bodies increase. Andcompared with group SR and group NS, the changes in group SA reducedsignificantly;3) The water content in brain tissues of each group began to increase after6hours in the middle third of the superior sagittal sinus and its reflux veins occlusion(compared with the group SO, P<0.05), and reached the peak at24hours afterligation, then decreased gradually with time prolonged, and at72hours, the watercontent significantly lower than24hours. Compared with the subgroup SR,thewater content of subgroup NS has no significant difference, but significantlyreduced as to the group SA (P<0.05);4) The expression of AQP-4protein of each group began to increase after6hours in the middle third of the superior sagittal sinus and its reflux veins occlusion(compared with the group SO, P<0.05), and reached the peak at24hours afterligation, then decreased gradually with time prolonged, and at72hours, the AQP-4protein significantly lower than24hours. Compared with the subgroup SR,theAQP-4protein of subgroup NS has no significant difference, but significantlyreduced as to the group SA (P<0.05).5) The content of EB of parasagittal brain tissue of each group began to increaseafter6hours in the middle third of the superior sagittal sinus and its reflux veinsocclusion (compared with the group SO, P<0.05), and reached the peak at24hoursafter ligation, then decreased gradually with time prolonged, and at72hours, thecontent of EB significantly lower than24hours. Compared with the subgroup SR,the EB content of subgroup NS has no significant difference, but significantlyreduced as to the group SA (P<0.05). 6) The brain water content was high positive linear correlated with the content ofEB(r=0.796,p<0.01), and the expression of AQP-4protein(r=0.773,p<0.01),thecontent of EB was high positive linear correlated with the expression of AQP-4protein(r=0.928,p<0.01).Conclusion:1) The brain water content, AQP-4protein and EB content areclosely related after the middle third of the superior sagittal sinus and its reflux veinsocclusion.2) Sodium aescinate can reduce the blood brain barrier by down-regulation ofAQP-4expression in the brain tissue to further reduce the degree of brain edema.
