ObjectiveTo observation the influence of peripheral blood white cells and mononuclear of rabbit bone-tendon junction damage under LIPUS treatment and to determine the best starting treatment timeMethodMethods148bone mature male Healthy New Zealand rabbits, undergone patellar tendon injury model, random divided into treatment group (n=100) and the control group (n=48), according to different starting time of postoperative treatment, the animals of treatment group were randomly divided into four subgroups:treatment group I, for the immediate postoperative LIPUS treatment group (n=48),treatment group Ⅱ, postoperative day3LIPUS treatment group (n=24), treatment group Ⅲ, for7days to begin treatment group (n=16) and treatment group Ⅳ,2weeks to begin treatment group (n=12). Animals as required in the postoperative respectively0h,4h,12h,24h and36h,3d,5d,1w,2w,4w and8w and16w to collect peripheral blood specimens and patellar tendon join complex (1weekago for postoperative hematoma surgical site), the routine blooddetected white blood cell and mononuclear cell; Flow Cytometry(FCM) and Immunohistochemistry detection CD163+.Results1. Detection of peripheral blood leukocytes1.1compared leukocyte count between difference groups:(1) at postoperative0h,4h,12h,24h and36h,3d and5d, statistically significant between treatment group I and control group (P<0.05);(2)at8w point, postoperative treatment group IV respectively were different between the treatment group Ⅱ, Ⅲ group (P<0.05);(3)at16w point, postoperative treatment group Ⅰ and group Ⅱ was statistically significant (P<0.05);1.2comparing the differences of the mononuclear cell count between groups:(1) at postoperative0h,4h,12h,24h and36h,3d and5d each point in time, there was statistically significant between treatment group Ⅰ and control group (P<0.05);(2) postoperative1w point, treatment group Ⅰ and group Ⅱ had significant difference (P<0.01);(3) at postoperative16w point, there was difference between the treatment group and control group IV (P<0.05);2. Flow cytometry test showed (1) there was no obvious difference between treatment group and control group at the same time;(2) there was no obvious difference between each point in time in the same group.3. The immunohistochemical detected CD163+. At16w point control group compared with each group (P<0.05).Immediate group compared respectively with3d,1w and2w group (P<0.05),1w and2w group (P<0.05);.According to the different time points within the same group compared:the control group,.immediate group,3d group,1w and2w groups were not statistically significants (P<0.05) after surgery1w,2w,4w and8w,and at16w time points after surgery1w,2w,4w and8w point had differences (P<0.05).ConclusionsAfter rabbit bone tendon junction injuried, LIPUS treatment had proinflammatory role in the early inflammatory response and had suppression inflammation effect in tissue repaire, and had the maximum therapeutic effect to begin after two weeks injuried.
