Objective:To disturb the hepatic fibrosis of rats induced by CCl14, with caffeine, and to record the change of relative index, what’s more, to discuss the therapeutic mechanism of caffeine.Methods:4groups was randomly devided for100SD rats. The concrete grouping was these ones:blank group (n=25), model group (n=25), caffeine treatment group (n=25) and caffeine control group (n=25). The rats of blank group accepted intraperitoneal injection of olive oil,3ml/kg of weight, twice a week for8weeks. The rats of model group and caffeine treatment group were given intralperitoneal injection of CCl4(CC14and olive oil1:1). what’s more, the dose and method were the same with blank group. The caffeine control group would not accept any management to be hepatic fibrosis model. At the same time, caffeine treatment group and caffeine control group were given caffeine50mg/kg administered once a day for8weeks, and the other groups were given CMC-Na (dose and method with the caffeine treatment group). In the9th week, made the HE, Masson staining to observe hepatic fibrosis semi-quantitative score of liver tissue, the liver inflammation semi-quantitative score, detect and calculate the expression of liver tissue’s a-SMA by staining of immunohistochemitry and with Real time qPCR, the expressing quantity of liver tissue TGF-β1CTGF and a-SMA.Results:At the end, the change of weight, pathological grading, the expression of TGF-β1, CTGF and a-SMA of model group, compared with the blank group, was significantly higher(P<0.01). Compared with model group, the change of weight, pathological grading, the expression of TGF-β1, CTGF of model group caffeine treatment group was also significantly higher(P<0.01). But the expression of a-SMA was relatively lower than the expression of TGF-β1and CTGF(P<0.05), which was statistically significant at the same time. Conclusion:(1)Caffeine can improve rat liver fibrosis induced by CC14to some extent.(2)With caffeine intaked constantly in a short time(8weeks), the liver of rats from caffeine control group has no significant difference with healthy rats (P>0.05).(3)The mechanism of caffeine which can treat the liver fibrosis may be as following: inhibiting the inflammation of the liver and the activation of hepatic stellate cell and myofibroblast, reducing the expression of promoting fibrosis factor TGF-β1and CTGF, which can unbalance the synthesis and degradation of ECM to promote the hepatic fibrosis.
