Objective:to verify whether there are interactions between FOLR1and Lyn、UBC、Ncaph2、IRA3、Cul3under physiological conditions.Method:both parental (CNE-1、HNE-2and5-8F) and Taxol-resistant NPC cells (CNE-1/Taxol、HNE-2/Taxol and5-8F/Taxol) were analyzed for protein interactions between FOLR1and the potential proteins(Lyn、UBC、Ncaph2、IRAK3、Cul3) by co-immunoprecipitation and Western blot using relevant antibodies.Result:1. The expression of Cul3、IRAK3and Ncaph2can be detected in the protein complexes of FOLR1initiated co-immunoprecipitation in the total protein of Taxol-resistant NPC cells (CNE-1/Taxol、HNE-2/Taxol and5-8F/Taxol) while that of Lyn and UBC9can not be detected. And all five antibodies were not detectable in the negative control group.2. The expression of FOLR1can be detected in the he protein complexes of co-immunoprecipitation of Cul3、IRAK3and Ncaph2antibodies with total protein of Taxol-resistant NPC cells (CNE-1/Taxol、 HNE-2/Taxol and5-8F/Taxol) but can not be detected in the complexes of Lyn and UBC9. and It was not detectable in NP69cells.Conclusion:1.There are interactions between FOLR1and Cul3、 IRAK3and Ncaph2in both parental and Taxol-resistant NPC cells. But there are no direct interactions between FLR1and Lyn or UBC9.2. Comparing with NP69cell lines, the expression of Cul3、IRAK3、 Ncaph2、 Lyn and UBC9were significantly increased in both parental and taxol-resistant NPC cell lines, and this may be associated with taxol-resistant nasopharyngeal carcinoma.
