Objective:To study the expression of interleukin-13(IL-13) and cyclin D1in airways and lungs of mice with asthma and the effect of airways smooth muscle hypertrophy on the expression, to investigate the effect of budesonide (BUD) and nicardipine (NI) in OVA-induced mice asthma models.Methods:Mice were sensitized and challenged with ovalbumin (OVA) to establish the asthmatic model. Sixty BALB/c SPF female mice were randomly divided into a control group, an asthma group, a BUD intervention group and a NI intervention group, with15mice in each group. To detect the pathological changes of each group mice airways and lungs tissue were observed by HE and Masson staining. The bronchial smooth muscle thickness (WAm/Pi) were measured by using computer-assisted image analysis system (Image-Pro Plus, IPP). The expression of interleukin-13(IL-13) and cyclin D1were detected by immunohistochemistry (IHC). The mRNA levels of interleukin-13(IL-13) and cyclin Dl was identified by RT-PCR.Results:1. Pulmonary histopathology:In asthma group, light microscopy of HE-stained airways and lungs tissue of mice were showed desquamation of airway epithelial cells, airway smooth muscle hypertrophy and sub-epithelial fibrosis. There was also presented much inflammatory cell infiltration around the airway and vessel. Masson stained lungs of asthma group demonstrated lots of blue collagen deposition around the airway and vessel. These changes were significantly reduced in BUD intervention group and NI intervention group.2. Computer-assisted image analysis:Compared with the control group (2.56±1.34), the WAm/Pi in bronchus were significantly higher in asthma group (9.12±2.46), BUD group (3.98±1.62) and NI group (6.32±1.59)(P<0.05). Compared with asthma group, BUD group and NI group were significantly decreased(P<0.05). Compared with NI group, BUD group were significantly decreased(P<0.005).3. IL-13immunohistochemical results:Compared with the control group(2.66±1.67)%, the protein expression of IL-13in asthma group (8.64±1.83)%and NI group(5.06±1.66)%were significantly increased (P<0.05). Compared with asthma group, BUD group and NI group were significantly decreased (P<0.05). Compared with NI group, BUD group were significantly decreased (P<0.05).4. Cyclin D1immunohistochemical results:Compared with the control group (7.24±2.23)%, the protein expression of Cyclin D1in asthma group(21.80v4.95)%, BUD group(10.60±3.88)%and NI group (14.64±5.48)%were significantly increased (P<0.05). Compared with asthma group, BUD group and NI group were significantly decreased (P<0.05). Compared with NI group, BUD group were significantly decreased (P<0.05).5. IL-13RT-PCR results:Compared with the control group(0.09±0.02), the expression of IL-13mRNA in asthma group(0.61±0.03), BUD group(0.17±0.02) and NI group(0.30±0.03) were significantly increased (P<0.05). Compared with asthma group, BUD group and NI group were significantly decreased(.P<0.05). Compared with NI group, BUD group were significantly decreased (P<0.05).6. Cyclin D1RT-PCR results:Compared with the control group(0.30±0.01), the expression of Cyclin D1mRNA in asthma group(1.30±0.01), BUD group(0.77±0.33) and NI group(0.89±0.01) were significantly increased (P<0.05). Compared with asthma group, BUD group and NI group were significantly decreased(P<0.05). Compared with NI group, BUD group were significantly decreased (P<0.05).7. Results of correlation:There was positive correlation between the protein expression of IL-13and Cyclin D1in asthma group (γ=0.702, P<0.05). There was positive correlation between the degree of airway smooth muscle proliferation and Cyclin D1expression (γ=0.915, P<0.05). There was positive correlation between the degree of airway smooth muscle proliferation and IL-13expression (γ=0.539, P<0.05).Conclusions:1.The protein and mRNA expression of IL-13and Cyclin D1in airway and lungs of mice with asthma significantly increased. There was positive correlation between the protein expression of IL-13and Cyclin D1. There was positive correlation between the degree of airway smooth muscle proliferation and Cyclin D1and IL-13expression.2. The protein and mRNA expression of IL-13and Cyclin D1in airway and lungs of mice decreased after Budesonide and Nicardipine treatment.3. The effect of Budesonide for asthma is better than Nicardipine.
