Objective:This study detect the level of miR-146a expression in the normal ovarian tissue, benign epithelial ovarian tumor, and epithelial ovarian carcinoma, and combined with clinical pathologic features to analyses the association of miR-146a expression with clinicopathological characteristics, to explore its significance in the development of epithelial ovarian carcinoma. Then we investigate the protein expression of IRAK1in the normal ovarian tissue and epithelial ovarian carcinoma,to explore the molecular mechanism of miR-186.We hope to reveal the pathogenesis of EOC and provide a new target for clinical diagnosis and therapy for EOC.Methods:1.The expression of miR-146a in the normal ovarian tissue, benign epithelial ovarian tumor,and epithelial ovarian carcinoma was examined by SYBR GreenI PCR, then analysis the association of miR-146a expression with clinicopathological characteristics in epithelial ovarian carcinoma.2.We predicted the downstream target genes of miR-146a from three databases and literature reports.The protein expression of IRAK1, a target genes of miR-146a in the normal ovarian tissue and epithelial ovarian carcinoma was examined by western blot analysis.Result:1.miR-146a expression was significantly higher in epithelial ovarian cancer samples compared with samples from normal ovary and benign epithelial ovarian tumor (P<0.05,P<0.05)2.miR-146a expression was significantly higher in poor-differentiated group than those in well-differentiated group group (P<0.05).There was no statistical difference in different age group、FIGO stage group、different CA125level group (P>0.05)3. IRAK1protein was low expressed in epithelial ovarian cancer samples which have a high level of miR-146a compared with samples from normal ovary (P<0.05).miR-146a and IRAK1protein’s expression significant correlation, and negatively related(r=-0.78, P=0.03).Conclusion:1.miR-146a expression was up-regulated in epithelial ovarian cancer tissue compared with samples from normal ovary and benign epithelial ovarian tumor.The up-regulation of miR-146a may play a significant role in EOC.2.miR-146a expression was significantly higher in poor differentiated group than those in well-differentiated group.miR-146a may correlated with tumor cell differentiation.3.IRAK1expression was significantly lower in epithelial ovarian cancer tissue than in normal ovary,miR-146a have significant negative correlation with IRAK1expression. IRAK1may be a target gene of miRNA-146a,miR-146a may promote the initiation and progression of EOC through IRAK1.
