Prognostic Implication Of The Expression Of Ezrin,Ki-67, Rb And TGF-β1in Soft Tissue Sarcomas

Objective:To clarify the correlation of the expression of Ezrin, Ki-67, Rb and TGF-β1with the clinicopathological variables and recurrence after operation in soft tissue sarcomas. Methods:Tissue microarrays from94soft tissue sarcomas patients were constructed from duplicate cores of viable and representative tumor areas.Immunohistochemistry from PV-6000polymer detection system was used to evaluate the correlation between the expression of Ezrin, Ki-67, Rb and TGF-β1, combined with the clinicopathological variables,and recurrence after operation from94soft tissue sarcomas patients.Results:Recurrence rate in the group of high expression of Ezrin in tumor was higher obviously,as well as Ki-67,and there was significant difference (P=0.026,0R=10.301;P=0.030,OR=8.467, respectively). While there was no statistical relationship between the expression of Rb and TGF-β1and recurrence of soft tissue sarcomas patients(P=0.544,P=0.367, respectively).But the expression of TGF-β1tended to correlate with gender (P=0.041). in addition, there was significantly difference between the different expression level of Rb and malignancy (P=0.030)-Proportion of the high expression of Rb in group of grade1or grade2((17.2%)) is apparently higher than that in group of grade3(2.8%)(P=0.030). In subgroup univariate analyses,the difference of recurrence rate in the group of different expression level of Ezrin and Ki-67between different size,different gender, different malignancy grade and different chemotherapy was statistically significant(P<0.05). however, In subgroup multivariate analyses, only the high expression of Ki67showed a positive correlation to recurrence rate in group of negative margin (P=0.029,OR=29.594).Conclusion:High expression of Ezrin, as well as Ki-67, in patients with soft tissue sarcomas and high expression of Ki-67in group of negative margin are independent predictors of recurrence.They could act as candidate markers,which contribute to preoperative evaluation and optimized treatment in patients with soft tissue sarcomas.
