Objective:Using pressing and solvent crystallization of horse fat unsaturated fatty acids for extraction, the main ingredients for oleic acid and linoleic acid, and adding antioxidants screening, development of horse fat unsaturated fattyacid soft capsule. Methods:1. Indexes to acid value, iodine value, peroxide value and yield, the single factor test, optimization of the extraction technology of horse crude oil.2. Using the solvent crystallization of horse fat unsaturated fatty acid extraction, the oil and gas chromatography mass spectrometry analysis of fatty acids in horse fat, the indexes to oleic acid yield as affected by a single factor test, select the best solvent and the optimum extraction times and the effects of crystallization temperature, crystallization time and mixed fatty acid and solvent ratio on horse fat unsaturated fatty acid solvent crystallization by orthogonal test.3. Horse fat unsaturated fatty acid as main component of soft capsule, was studied by single factor method for screening antioxidants of type and adding amount, determine the soft capsule preparation process. Results:1. Pressing effect of different extraction time method factors greater than the effect of different extraction temperature; the optimal extraction process was as follows:extraction time6h,extraction temperature80℃; this condition data is the most ideal results, the yield is39.44±0.07g, iodine value, peroxide value and acid value were3.580mg/g,81.012g/100g,0.916mmol/kg.2. Solvent extraction process:obtained by single factor test:the best solvent was95%ethanol, the optimum process conditions is the crystallization temperature of2℃, the best crystallization time6h,1:7is the best mixed fatty acid and solvent ratio; optimal number of filter cake repeated crystallization for two times; by orthogonal test, the optimum technological conditions are:1:9is mixed fatty acid and solvent ratio; crystallization time was8h; crystallization temperature of2℃.3.The best kind of antioxidant is vitamin E, the optimum adding amount was0.02%, the best ratio of gelatin glycerin water soft capsule shells for1:0.5:1. Conclusion:Low temperature pressing relative solvent extraction method is more convenient, safe, and high yield, is suitable for large-scale industrial production; solvent crystallization method after extraction of unsaturated fatty acid content was extracted before increased24.39%, oleic acid content increased by20.21%. The solvent crystallization method after extraction of horse oil, containing more unsaturated fatty acids, can be developed into health care products potential.
