Objective:To evaluate the effect of Yili dark bee propolis on the main cariogenic bacterium and biofilm and to find the mechanism.Methods:①Susceptibilities to the ethanolic extract of propolis (EEP) against S.mutans, S.sobrinus,S. sanguis, L.acidophilus, A.viscosus and A.naeslundii were analyzed by the liquid diffusion method and determining the minimum inhibitory concentration (MIC) and minimum bactericidal concentration (MBC);②Susceptibilities to the ethanolic extract of propolis (EEP) against S.mutans, S. sobrinus,S. sanguis,A.viscosus and A.naeslundii were analyzed by the Crystal Violet Stain method and determining the minimum biofilm eradication concentration (MBEC);③The concentration of control groups are lower than MIC with initial pH7.0and vacuity contrast group cultured for48h,then measure their pH to evaluate acid producing ability of tested bacteria;④Evaluate the effect of propolis on extracellular polysaccharides synthesis of S.mutans and A.viscosus by anthrone method;⑤Cultivating24h biofilm first,then the control groups with concentration of lower than MBEC which initial pH7.0and vacuity contrast group cultured for24h,then measure their pH to evaluate acid producing ability of tested plaque biofilm;⑥Evaluate the effect of propolis on insoluble extracellular polysaccharides synthesis of S.mutans biofilm by anthrone method.Results:①The MIC of Yili propolis on S.mutans,S. sobrinus,S. sanguis, L.acidophilus, A.viscosus and A.naeslundii were0.78mg·mL-1、0.39mg·mL-1、1.56mg·mL-1、0.39mg·mL-1、0.2mg·mL-1、0.2mg·mL-1respectively; The MBC of Yili dark bee propolis on tested bacteria were1.56mg·mL-1'0.78mg·mL-1、3.125mg·mL-1、0.78mg·mL-1、0.39mg·mL-1、0.39mg·mL-1;②The MBEC of Yili propolis on S.mutans,S. sobrinus,S. sanguis,A.viscosus and A.naeslundii were6.25mg·mL-1、1.56mg·mL-1、3.1256mg·mL-1、0.78mg·mL-1、0.78mg·mL-1respectively;③propolis could decrease the△pH of tested bacteria,the difference between control groups and vacuity contrast group were statistically significant(P<0.05);④At MIC concentrate,propolis could reduce the ability of S.m and A.viscosus synthtic extracellular polysaccharides,the difference were statistically significant(P<0.05);⑤propolis could decrease the ApH of tested plaque biofilm,the difference between control groups and vacuity contrast group were statistically significant(P<0.05);⑥At MBEC concentrate,propolis could reduce the ability of S.m synthtic insoluble extracellular polysaccharides,the difference were statistically significant(P<0.05).Conclusion:Yili propolis is a kind of nature products showed not only remarkable antimicrobial activities against the cariogenic oral bacteria and biofilm and have effect on anticaries.
