Objective:1.To observe the changes of serum DKK1level and Peripheral Blood Mononuclear Cells(PBMC) DKK1mRNA expression in ankylosing spondylitis(AS) patients before and after Glucoside Tripterygium Total(GTT) treatment;2.To explore the relevancy of serum DKK1level and PBMC DKK1mRNA expression during AS development;3.To evaluate effectiveness and the usage safety of GTT in AS.Methods:This test adopts self-reflection and blank control method..40patients were treated with GTT in experimental group. Serum DKK1level and PBMC DKK1mRNA expression of subjects before and after treatment were recorded in detail.Recorded the primary endpoint (BASDAI,BASMI, BASFI,ASAS20/40) and secondary efficacy endpoint(ESR, CRP) for AS patients,and analyzed the correlation between them. Monitored and recorded the adverse drug reactions with accordingly recording.Results:1.Before treatment, serum DKK1level in the experimental group are lower than the blank control group,with significant difference (P<0.01);2.Serum DKK1level and PBMC DKK1mRNA expression in the experimental group had a significant difference before treatment(P<0.01);3.GTT has a good effect in keeping improving primary indicators(BASDAI, BASMI, BASFI,ASAS20/40)and secondary indicators(ESR, CRP)in the experimental group;4.Serum DKK1level and PBMC DKK1mRNA expression had no correlation with all of every indicators for AS patients;5.Fourteen cases of adverse reactions appeared in experimental group (8cases of gastrointestinal discomfort,4cases of liver damage,and2cases of reproductive system damage),improved by dose reduction or discontinuation and symptomatic treatment.Conclusion:1.serum DKK1level of AS patients with active period are significantly lower than healthy people;2.After treated by GTT, serum DKK1level and PBMC DKK1mRNA expression of AS patients are elevating than before;3.GTT can possible elevating serum DKK1level and PBMC DKK1mRNA expression;4.GTT shows good effect for treating AS in a short time, and relatively safe by using in small doses under close monitoring.
