Objective:To explore the anti-carcinoma of lung and lung cancer cells’ function mechanism of ruscogenin, which is monomer of Radix Ophiopogonis in zhou’s prescription from Chinese national expert Zhou Zhongying’s experience prescriptions, so as to illustrate the molecular biological basis for the "cancer toxin" theory proposed by Prof. Zhou, providing evidence for its clinical application and theoretical basis for the research and production of new anti-tumor drugs.Methods:Human lung cancer cells A549and H460were cultured in vitro. The effects of ruscogenin on lung cancer cells’growth, they are from experimental groups were detected by MTT method, and cisplatin as positive control group. Flow cytometer was applyed to observe the influence of cell apoptosis rate and cell cycle distribution. Western Blot were applied to find out the effects of ruscogenin on cell cycle’s relevant protein. Transwell is used to realize the cells’ invading ability.Results:MTT showed that ruscogenin of12.5~100umol/L had significant effects of inhibiting lung cancer cells’proliferation in an evident time and dose dependence manner, there are obviously difference compare to the control group. FCM showed that after24h, cell apoptosis began and increased along with the increased ruscogenin dose relative to the control group, ratio of cells at S phase increased comparatively and cells of G0/G1phase reduced, explain the cell cycle was blocked at S phase. Western Blot detection indicated that p53and p21protein expression increased, but CDK2and CyclinE2protein decreased. The Transwell manifested that ruscogenin can inhibit the lung cancer cells’invading ability.Conclusion:Ruscogenin is one active ingredient in Radix Ophiopogonis, which can significantly inhibit human adenocarcioma A549and H460cells’ proliferation, it’s main mechanism might be related to its inhibition of S phase, with the p53and p21protein expression increased, CDK2and cyclinE2protein expression decreased. The study shows that p53and p21protein were highly expressed,CDK2and cycclinE2protein low expression can be one of the mechanism of its effects.
