Objective: To observe the changes in nerve function recovery, themorphology of hippocampal neurons and the expression of Reelin, Dab-1,PI3KCA in cerebral ischemic rats after treadmill running exercise.Methods: Male Sprague-Dawley rats were randomly divided intothree groups: sham-operated group (S group), ischemic control group (Igroup) and treadmill running exercise group (E group). Middle cerebralartery occlusion (MCAO) was used to create focal cerebral ischemicmodels. Rats in E group underwent treadmill running exercise for16days,(15m/min,30min/d). Triphenyltetrazolium chloride staining (TTCstaining) was used to observe the cerebral infarction of rats after theMCAO operation. Modified neurological severity scores (mNSS) was usedto evaluate the neurological function of rats in three groups. HE stainingwas used to observe the morphology of hippocampal neurons of rats inthree groups. Western blot was used to detect the expression of Reelin andDab-1and immunohistochemistry was used to detect the number ofReelin-positive and PI3KCA-positive cells in three groups. Result: The mNSS of E group was lower than that of I group at day19post-MCAO (P<0.05). The morphology of the hippocampal neurons inS group was normal, irregular and damaged in I group, while alleviatedafter treadmill running exercise. The expression of Reelin in ischemiccortexs of rats in E group was remarkably upregulated than that in S and Igroup with statistical differences(P<0.05). The expression of Dab-1inischemic cortexs of rats in E group was remarkably downregulated thanthat in S and I group with statistical differences(P<0.05). The expression ofReelin-positive cells in E group were higher than that in I group(18.27±4.03vs.7.53±2.03, P<0.01) and the PI3KCA-positive cells in Egroup were also higher than that in I group (10.27±1.62vs.5.40±1.84, P<0.01).Conclusion: Treadmill running exercise can improve neurologicalfunction, maintain the relatively normal morphology of the hippocampalneurons and change the expression of Reelin, Dab-1,PI3KCA in rats aftercerebral ischemia, which is possibly involved in the molecular and cellularrepair mechanisms of treadmill running exercise after stroke.
