Objective: The aim of the present study is to investigate the influenceof loading exercise on morphology and metabolism of articular cartilagein ovariectomized rats.Methods: Fifty female Sprague-Dawley(SD)rats were randomlyassigned to5groups, after successfully preparation of osteoporosismodel: ovariectomized (OVX), ovariectomized plus exercise(OVX-RUN), ovariectomized plus low loading exercise (low loadingOVX-RUN), ovariectomized plus medium loading exercise (mediumOVX-RUN), ovariectomized plus high loading exercise (high loadingOVX-RUN); After12weeks, rats were killed and the followingvariables were compared among the5groups. Knee joint and bloodsamples were collected. Test of rat serum related material were carriedon the articular cartilage morphology: Hematoxylin and Eosin (HE)dyeing, sarranine O(SO) dyeing, toluidine dyeing examination andimmunohistochemical staining to detect the content of type â…¡collagen in articular cartilage.Results: Morphological tests revealed that medium OVX-RUN groupwas deeper in Hematoxylin and Eosin (HE) dyeing, sarranine O(SO)dyeing, toluidine dyeing,compared with OVX〠OVX-RUNã€lowloading OVX-RUN〠high loading OVX-RUN groupï¼› Rats inovariectomized plus high loading exercise (high loading OVX-RUN)had significantly higher Mankin′s score and express of MMP–3thanovariectomized (OVX), ovariectomized plus exercise (OVX-RUN),ovariectomized plus low loading exercise (low loading OVX-RUN),ovariectomized plus medium loading exercise (medium OVX-RUN)(P<0.05), type â…¡ collagen had significantly lower than ovariectomized(OVX), ovariectomized plus exercise (OVX-RUN), ovariectomized pluslow loading exercise (low loading OVX-RUN), ovariectomized plusmedium loading exercise (medium OVX-RUN)(P<0.05);ovariectomized plus medium loading exercise (medium OVX-RUN) hadsignificantly Mankin′s score and expression of MMP–3thanovariectomized (OVX), ovariectomized plus exercise (OVX-RUN),ovariectomized plus low loading exercise (low loading OVX-RUN),ovariectomized plus high loading exercise (high loading OVX-RUN)ï¼›type â…¡collagen had significantly higher than ovariectomized (OVX),ovariectomized plus exercise (OVX-RUN), ovariectomized plus lowloading exercise (low loading OVX-RUN), ovariectomized plus high loading exercise (medium OVX-RUN)(P<0.05).Conclusion: Knee joint cartilage of rats presented different changeunder the different load movement. Low loading exercise hadunobvious effect on joint cartilage, high loading not only cause thedegenerative diseases of cartilage in rat,but also cause expression ofMMP–3rise in blood, destroy the structure of collagen andproteoglycan, thus cause cartilage injuries. Medium loading exercise ishelpful to maintain the steady state of articular cartilage,accroding to themorphology and serum test results. Therefore,moderate loading exerciseon the prevention and treatment of osteoporosis had better useful clinicalsignificance. |
