Purpose: The present study was performed to investigate theconcentrations of the H2S in plasma and vitreous with diabetic retinopathypatients,to discuss the relative levels of VEGF and H2S in plasma andvitreous with diabetic retinopathy patients.Methods:Blood samples were collected from normal controls anddiabetic patients. Vitreous samples were collected from patients withproliferative diabetic retinopathy (PDR) and patients with rhegmatogenousretinal detachment (RRD). Diabetic patients were grouped into diabeticpatients without diabetic retinopathy (non-DR), diabetic patients withnon-proliferative DR (NPDR) and patients with PDR. Concentrations ofH2S and vascular endothelial growth factor (VEGF) in the plasma andvitreous body were detected using a spectrophotometer.Results:A decreased H2S level in the plasma was observed in non-DRgroup (41.89±8.52μM, p<0.05) and an increased H2S level in the plasmawas PDR group (60.49±11.14μM, p<0.01), when compared with that innormal controls (49.67±9.72μM). There was no difference in plasma H2S Level between patients with NPDR (54.13±8.61μM) and normalcontrols(p=0.53). In the vitreous body, H2S levels in PDR group weresignificantly higher (76.80±24.08μM, p<0.01) than that in RRD group(24.37±11.25μM). Levels of VEGF in plasma from diabetic patients weresignificantly lower (p<0.01) than that in normal controls. VEGF levels inthe vitreous body from diabetic patients with PDR were significantlyhigher (885.61±190.41pg/ml, p<0.01) than that from patients with RRD(89.98±19.56pg/ml). Seven days after an intravitreal injection ofranibizumab, a significantly decreased H2S level (55.58±7.20μM, p<0.05)was observed in the vitreous body in patients with PDR, when comparedwith that (75.07±12.95μM) in the vitreous body collected just beforeintravitreal injection.Conclusions: These results indicated that anti-VEGF maydownregulate the H2S level in the vitreous body and H2S may play a role inthe development of DR. H2S may be a novel target for the therapy of DR.
